摘要:
目的 探讨两种乙型肝炎病毒DNA试剂检测HBV-DNA的相关性,分析其检测结果的差异性。方法 使用凯杰试剂与之江试剂检测75例患者血清中的HBV-DNA水平,凯杰试剂采用手工提取样本中的DNA,之江试剂采用自动核酸提取仪提取DNA,然后进行核酸扩增。对HBV-DNA病毒载量进行对数转换(lg10),并将两组数据进行相关性分析;对不一致的结果采用罗氏试剂进行确证。结果 以HBV-DNA含量>5.0×102 IU/ml临界值,拟合两组检测数据结果,相关方程为Y=0.8889X+0.7956(R2=0.8954,P<0.05);凯杰试剂检测标本中HBV-DNA含量为[(5.07±1.96)(对数值)],之江试剂检测结果为[(5.30±1.84)(对数值)],差异有统计学意义(P<0.05)。凯杰试剂检测阳性率为57.33%(43/75),低于之江试剂的73.33%(55/75),差异有统计学意义(P<0.05)。15例检测结果不一致的样本中,凯杰试剂与罗氏试剂结果符合率为37.50%,之江试剂与罗氏试剂检测结果符合率为62.50%。结论 凯杰试剂与之江试剂相关性良好,其检测结果可以比较,且之江试剂检测HBV-DNA的敏感性高于凯杰试剂。
Abstract:
Objective To investigate the correlation between two hepatitis B virus DNA reagents for detecting HBV-DNA and analyze the differences in their detection results. Methods The levels of HBV-DNA in the serum of 75 patients were detected by using Kaijie reagent and Zhijiang reagent. The Kaijie reagent used manual extraction of DNA from the sample. The Zhijiang reagent used an automatic nucleic acid extractor to extract DNA and then perform nucleic acid amplification. The HBV-DNA viral load was log-transformed (lg10), and the two sets of data were correlated. The inconsistent results were confirmed by Roche reagent.Results The HBV-DNA content>5.0×102 IU/ml threshold value was used to fit the results of the two groups. The correlation equation was Y=0.8889X+0.7956 (R2=0.8954, P<0.05).The HBV-DNA content in the Kaijie reagent test specimens was [(5.07±1.96) (logarithmic value)], and the Zhijiang reagent test result was [(5.30±1.84) (logarithmic value)], the difference was statistically significant (P<0.05). The positive rate of Kaijie reagent detection was 57.33%(43/75), which was lower than that of Zhijiang reagent 73.33%(55/75).The difference was statistically significant (P<0.05). Among the 15 samples with inconsistent test results, the coincidence rate of Kaijie reagent and Roche reagent was 37.50%, and the coincidence rate between Zhijiang reagent and Roche reagent was 62.50%.Conclusion Compared with the two reagents, the correlation is good and the test results can be compared. The sensitivity of Zhijiang reagent to detect HBV-DNA is higher than that of Kaijie reagent.
