摘要:
目的 研究糖尿病大鼠心肌损伤对血清瘦素水平的影响。方法 采用随机数字表法将50只健康大鼠分为正常对照组16只,模型组34只。正常组大鼠给予普通饲料喂养,模型组给予高糖高脂饲料喂养建立糖尿病大鼠模型,喂养8周后,腹腔注射STZ形成糖尿病模型后,继续给予高糖高脂喂养12周,依据是否合并心肌病分为糖尿病组(T2DM组)16只和糖尿病心肌病组(DCM组)12只,比较各组大鼠心脏功能指标、血糖、体重、血脂指标、血清瘦素水平、血浆脑钠肽(BNP)水平,分析血清瘦素水平与糖尿病大鼠心肌病的关系。结果 DCM组E/A比值大于正常对照组、T2DM组,LVEF小于正常对照组、T2DM组,差异有统计学意义(P<0.05);T2DM组与正常对照组比较,差异无统计学意义(P>0.05);三组大鼠体重、胆固醇比较,差异无统计学意义(P>0.05);T2DM组、DCM组血糖、甘油三酯水平均大于正常对照组,差异有统计学意义(P<0.05),但T2DM组、DCM组血糖比较,差异无统计学意义(P>0.05);T2DM组、DCM组血清瘦素、BNP水平均大于正常对照组,且DCM组大于T2DM组,差异有统计学意义(P<0.05)。结论 血清瘦素与糖尿病大鼠心肌病的发生、发展密切相关,其水平下降可一定程度改善糖尿病大鼠心肌病预后,减轻心肌损伤。
Abstract:
Objective To study the effect of myocardial injury on serum leptin level in diabetic rats.Methods Fifty healthy rats were randomly divided into normal control group (n=16) and model group (n=34), the normal control group was fed with ordinary diet, while the model group was fed with high-glucose and high-fat diet to establish diabetic rat model for 8 weeks, when the STZ-injected rats model was established, and then fed with high-glucose and high-fat diet for 12 weeks, according to whether or not the cardiomyopathy was divided into diabetic group (T2DM group, n=16) and diabetic cardiomyopathy group (DCM group, n=12). The heart function index, blood sugar, body weight, blood fat index, serum leptin level and plasma brain natriuretic peptide (BNP) level were compared in each group and the relationship between serum leptin level and cardiomyopathy in diabetic rats was analyzed.Results The E/A ratio in DCM group was higher than that in normal control group, T2DM group, while LVEF was lower than that in normal control group, T2DM group, and the difference was statistically significant (P<0.05), there was no significant difference between T2DM group and normal control group (P>0.05); there was no significant difference in body weight and cholesterol among the three groups (P>0.05); the levels of blood glucose and triglyceride in T2DM group and DCM group were higher than those in normal control group, and the difference was statistically significant (P<0.05),but there was no significant difference in blood glucose between T2DM group and DCM group (P>0.05); the levels of serum leptin and BNP in T2DM group and DCM group were higher than those in normal control group, and DCM group was higher than that in T2DM group, nd the difference was statistically significant (P<0.05).Conclusion Serum leptin is closely related to the occurrence and development of diabetic cardiomyopathy, and its decrease may improve the prognosis of diabetic cardiomyopathy to some extent, reduce myocardial injury.
