摘要:
目的 研究乳头状甲状腺癌和癌旁组织中ACACB基因mRNA和蛋白表达水平与ACACB基因启动子甲基化水平的相关性。方法 于2020年11月-2021年3月在云南省第一人民医院采集10例乳头状甲状腺癌患者的肿瘤组织和癌旁正常组织样本,用qRT-PCR和WB分别检测10对乳头状甲状腺癌和对应癌旁组织中ACACB基因mRNA和蛋白水平,使用甲基化引物通过PCR检测ACACB基因是否发生启动子甲基化。结果 乳头状甲状腺癌组织中ACACB基因mRNA水平均低于癌旁正常组织,但不同患者的差异程度有所不同;8例患者ACACB基因蛋白水平低于癌旁正常组织,2例患者ACACB基因的蛋白表达水平高于癌旁正常组织;6例患者肿瘤组织中ACACB基因发生启动子甲基化,3例患者癌旁正常组织中ACACB基因发生启动子甲基化,1例患者肿瘤组织与癌旁正常组织同时发生甲基化。结论 乳头状甲状腺癌中ACACB启动子甲基化会影响其转录活性,进而下调乳头状甲状腺癌中ACACB蛋白的表达,但其具体的作用机制尚不明确。
Abstract:
Objective To investigate the correlation between mRNA and protein expression of ACACB gene and promoter methylation of ACACB gene in papillary thyroid carcinoma and paracancerous tissues.Methods From November 2020 to March 2021, cancer and paracancerous samples were collected during tumor resection in the First People’s Hospital of Yunnan Province. The mRNA and protein levels of ACACB gene in 10 pairs of papillary thyroid carcinoma and corresponding paracancerous tissues were detected by qRT-PCR and WB, respectively. Methylation primers were used to detect promoter methylation of ACACB gene by PCR.Results The mRNA level of ACACB gene in all papillary thyroid carcinoma tissues was lower than that in adjacent tissues, but the degree of difference was different in tissues of different patient origin. The protein level of ACACB gene in 8 patients was lower than that in adjacent normal tissues, and the protein expression level of ACACB gene in 2 patients was higher than that in adjacent normal tissues. ACACB gene promoter methylation occurred in 6 cases of tumor tissues, 3 cases of adjacent normal tissues and 1 case of tumor tissues and adjacent normal tissues.Conclusions ACACB promoter methylation in papillary thyroid cancer can affect its transcriptional activity and thus down-regulate the down-regulation of ACACB protein in papillary thyroid carcinoma, but the specific mechanism remains unclear.
