摘要:
目的 研究杠香乙醇提取物对小鼠的急性毒性反应情况,为杠香的临床药用和后续毒理药效研究提供参考。方法 取SPF级昆明种小鼠20只,雌雄各半,随机分为空白组和杠香组,每组10只,雌雄各5只。采用14 d最大给药量法进行小鼠急性毒性实验,杠香组给予小鼠一次性灌胃最大浓度杠香乙醇提取物,空白组给予等量蒸馏水;给药后连续14 d观察小鼠生理反应及行为状态,记录两组小鼠体重变化和进食情况;14 d后解剖小鼠,观察脏器变化,取其脏器切片,HE染色,行组织病理学检查。结果 杠香组小鼠出现活动减少,喜静卧,1 h内全部恢复正常,其他观察指征无明显变化;实验14 d内,两组体重、脏器系数比较,差异无统计学意义(P>0.05),但杠香组雌性小鼠给药前后进食量整体较低,雄性小鼠给药前后进食量整体偏高;剖检肉眼观察两组小鼠的肝、心、脾、肺、肾未发现明显损伤情况,组织病理学观察无异常。结论 杠香乙醇提取物未见明显毒性,临床使用安全性较高。
Abstract:
Objective To study the acute toxicity test of ethanol extract of Gangxiang in mice, and to provide reference for clinical medicine and subsequent toxicological and pharmacodynamic studies of Gangxiang.Methods Twenty SPF grade Kunming mice, half female and half male, were randomly divided into blank group and Gangxiang group, with 10 mice in each group, 5 females and 5 males each. The acute toxicity test of mice was carried out by 14 d maximum dose method. The mice in the control group were given the same amount of distilled water, while those in the experimental group were given the maximum concentration of ethanol extract. After 14 consecutive days of administration, the physiological response and behavioral status of mice were observed, and the weight changes and eating conditions of mice in the two groups were recorded. After 14 days, the mice were dissected, the changes of organs were observed, the organs were sliced, stained with HE, and histopathological examination was performed.Results The mice in the Gangxiang group showed decreased activity and preferred to lie still, all of which returned to normal within 1 h, and other observation indications did not change significantly. Within 14 days of the experiment, there was no significant difference in body weight and organ coefficient between the two groups (P>0.05). However, the food intake of female mice in the Periploca sepium group was lower before and after administration, and the food intake of male mice was higher before and after administration. No obvious damage was found in the liver, heart, spleen, lung and kidney of the two groups of mice, and there was no abnormality in histopathological observation.Conclusion There is no obvious toxicity in the ethanol extract of Gangxiang, and it is safe for clinical use.
