摘要:
目的 探讨血型血清学方法鉴定ABO疑难血型,结合分子生物学方法准确定型。方法 收集2019年8月-2021年7月本血站检验科送检的ABO疑难血型标本26例,对ABO疑难血型标本采用血型血清学方法、聚合酶链式反应-序列特异性引物法(PCR-SSP)及基因测序(PCR-SBT)技术进行鉴定。结果 19例标本存在不规则抗体,以抗M抗体为主;另外7例疑似ABO亚型标本经PCR-SSP基因分型检出1例A307/O02 亚型,对其中6例标本PCR-SSP未明确血型通过测序确定基因型,2例标本存在O01/O02/B 3个等位基因,属血型嵌合体, 2例B101/O01,1例A102/O02,1例B101/O02。结论 鉴定献血者ABO疑难血型时,应用血清学和基因检测及DNA测序的方法,有助于正确鉴定血型。
Abstract:
Objective To investigate the serological method for identification of difficult ABO blood group and the molecular biological method for accurate typing.Methods A total of 26 difficult ABO blood group samples sent by the Laboratory Department of our blood station from August 2019 to July 2021 were collected. The difficult ABO blood group samples were identified by blood group serology, polymerase chain reaction sequence specific primer (PCR-SSP) and gene sequencing (PCR-SBT).Results There were irregular antibodies in 19 samples, mainly anti-M antibody. In addition, one A307/O02 subtype was detected in 7 suspected ABO subtype samples by PCR-SSP genotyping. The genotypes of 6 samples were determined by sequencing, while there were 3 alleles of O01/O02/B in 2 samples, belonging to blood group chimera, 2 cases of B101/O01, 1 case of A102/O02 and 1 case of B101/O02.Conclusion When identifying difficult ABO blood groups of blood donors, the methods of serology, gene detection and DNA sequencing can correctly identify blood groups.
