摘要:
目的 运用生物信息学分析方法分析长链非编码RNA(CASC9)在细胞内的调控靶基因及信号通路,并对其进行生物学功能注释,阐述CASC9在细胞内的功能。方法 在miRDB数据库和Starbase3.0数据库中预测与CASC9有miRNA-lncRNA互作关系的miRNA,将结果取交集;在Starbase3.0数据库中预测miRNAs靶基因,并进行GO功能注释和Pathway通路富集。结果 miRDB数据库预测到50个miRNA与CASC9相作用,Starbase数据库预测到23个miRNA与之相作用,取交集共得当16个miRNA;GO功能注释显示16个miRNA的靶基因主要分布在细胞内金属离子功能,如镁离子等;生物学过程主要分布在蛋白质的磷酸化和信号通路方面;信号通路富集结果主要分布在泛素介导的蛋白质降解通路、Autophage相关信号通路、MAPK信号通路、RAS信号通路以及细胞内的肿瘤信号通路等。结论 长链非编码RNA CASC9在细胞内的调控作用与镁离子结合、蛋白质磷酸化调控、蛋白质泛素化降解、自噬以及多种肿瘤发生信号通路密切相关,其可通过上述作用通路调控疾病的病理进展。
Abstract:
Objective To analyze the regulatory target genes and signaling pathways of long non-coding RNA ( CASC9 ) in cells by bioinformatics analysis, and to annotate its biological functions, and to explain the function of CASC9 in cells.Methods The miRNAs with miRNA-lncRNA interaction with CASC9 were predicted in miRDB database and Starbase3.0 database, and the results were intersected. Target genes of miRNAs were predicted in the Starbase 3.0 database, and GO functional annotation and Pathway pathway enrichment were performed.Results A total of 50 miRNAs were predicted to interact with CASC9 in the miRDB database, and 23 miRNAs were predicted to interact with CASC9 in the Starbase database. GO functional annotation showed that the target genes of 16 miRNAs were mainly distributed in intracellular metal ion functions, such as magnesium ions. Biological processes were mainly distributed in protein phosphorylation and signaling pathways; the enrichment results of signaling pathway were mainly distributed in ubiquitin-mediated protein degradation pathway, Autophage-related signaling pathway, MAPK signaling pathway, RAS signaling pathway and intracellular tumor signaling pathway.Conclusion The regulation of long non-coding RNA CASC9 in cells is closely related to magnesium ion binding, protein phosphorylation regulation, protein ubiquitination degradation, autophagy and various tumor signaling pathways, which can regulate the pathological progress of diseases through the above pathways.
