摘要:
目的 研制治疗急性踝关节扭伤的大黄祛伤巴布剂。方法 ①以溶剂分数、倍量、提取时间、提取次数作为主要考察因素,以高效液相色谱法检测大黄总蒽醌、栀子苷、芍药苷含量,并作为考察指标,采用单因素分析法优化提取工艺;②以聚丙烯酸钠NP-700、甘羟铝、甘油作为3个主要考察因素,以剥离性、涂展性、均匀性为考察指标进行L9(34)正交试验优化制备工艺。结果 ①最佳提取工艺为:除乳香、没药和樟脑外其余成分在90 ℃条件下,70%乙醇,8倍量,加热回流2次,第1次2 h,第2次1 h;乳香、没药在常温条件下,乙醇,10倍量,静置24 h;樟脑直接碾碎加入甘油项;②最优基质配方为:聚丙烯酸钠NP-700∶甘羟铝∶甘油=5∶0.25∶40。结论 按最优提取方法、最优基质配比制得的大黄祛伤巴布剂,其剥离性、涂展性、均匀性良好,制备工艺稳定可行。
Abstract:
Objective To study the preparation technology of Dahuang Qushang cataplasm.Methods ①The contents of total anthraquinones, geniposide and paeoniflorin in rhubarb were determined by high performance liquid chromatography with solvent fraction, double amount, extraction time and extraction times as the main factors. ②The extraction process was optimized by single factor analysis. L9(34) orthogonal test was used to optimize the preparation process with sodium polyacrylate NP-700, aluminum glycinate and glycerol as the three main factors, and peelability, spreadability and uniformity as the indexes.Results ①The optimal extraction technology was as follows: the preparation with 8 times 70% ethanol boiled out twice in 90℃, the first time for 2 hours, and the second time for 1 hours. The optimal extraction technology of Olibanum and Myrrha were to extract once with 10 times of 95% ethanol for 24 hours. ②The optimal matrix formulation of Dahuang Qushang cataplasm was viscomate NP-700∶aluminium glycinate∶glycerol=5∶0.25∶40.Conclusion Dahuang Qushang cataplasm was made according to the optimum extraction process and matrix formula of the system, and its repeated peeling flexibility and adhesion to the skin meet the requirement of cataplasm.
