摘要:
目的 观察归芪益元膏对气虚证小鼠的干预作用。方法 采用力竭游泳法复制出气虚证模型小鼠。实验动物随机分为5组,每组10只。空白组和模型组灌胃等体积蒸馏水,归芪益元膏低、中、高剂量组分别给予归芪益元膏1.6、3.3、6.6 g/kg。比较给药前后各组小鼠全血白细胞(WBC)、红细胞(RBC)、血红蛋白(Hb)、血小板(PLT)数目的变化,取脾脏、胸腺,对其称重,计算脾脏指数(SI)、胸腺指数(TI)的变化,各组小鼠力竭游泳时间、归芪益元膏对气虚小鼠肾上腺素及去甲肾上腺素含量、免疫细胞、肝糖原及肌糖原含量、胸腺指数、脾脏指数的影响。结果 与空白组比较,模型组外周WBC、RBC、Hb、PLT含量显著降低(P<0.01),归芪益元膏中、高剂量组外周WBC、RBC、Hb、PLT含量升高(P<0.05);与空白组比较,模型组小鼠力竭游泳时间缩短(P<0.05);与模型组比较,归芪益元膏高、中、低剂量组小鼠力竭游泳时间增加(P<0.05);与空白组比较,模型组小鼠肾上腺素及去甲肾上腺素浓度均有所降低(P<0.01);与模型组比较,归芪益元膏高、中剂量组肾上腺素及去甲肾上腺素浓度均有明显升高(P<0.01);与空白组比较,模型组B细胞、T淋巴细胞亚群CD4+、CD8+水平均显著降低(P<0.01);与模型组比较,归芪益元膏高剂量组T 淋巴细胞亚群CD4+、CD8+水平均显著升高;与空白组比较,模型组小鼠肝糖原、肌糖原含量、脾脏指数、胸腺指数显著降低(P<0.01);与模型组比较,归芪益元膏高、中、低剂量组大鼠肝糖原、肌糖原含量、脾脏指数显著增加(P<0.01);与模型组比较,归芪益元膏高、中、低剂量组胸腺指数升高(P<0.05)。结论 归芪益元膏可以明显改善力竭游泳所致气虚小鼠的运化及免疫功能,其中高、中剂量组效果更加明显。
Abstract:
Objective To observe the effect of Guiqi Yiyuan ointment on mice with Qi deficiency syndrome.Methods The model mice of Qi deficiency syndrome were established by exhaustive swimming. The experimental animals were randomly divided into 5 groups, with 10 mice in each group. The blank group and the model group were given an equal volume of distilled water, and the low, medium and high dose groups of Guiqi Yiyuan ointment were given Guiqi Yiyuan ointment 1.6, 3.3 and 6.6 g/kg, respectively. The changes of white blood cell (WBC), red blood cell (RBC), hemoglobin (Hb) and platelet (PLT) in whole blood of mice in each group before and after administration were compared. The spleen and thymus were taken and weighed, and the changes of spleen index (SI) and thymus index (TI) were calculated. The exhaustive swimming time of mice in each group and the effects of Guiqi Yiyuan ointment on adrenaline and norepinephrine content, immune cells, liver glycogen and muscle glycogen content, thymus index and spleen index of Qi deficiency mice were compared.Results Compared with the blank group, the contents of WBC, RBC, Hb and PLT in the peripheral blood of the model group were significantly decreased(P<0.01), and the contents of WBC, RBC, Hb and PLT in the middle and high dose groups of Guiqi Yiyuan ointment were increased (P<0.05). Compared with the blank group, the exhaustive swimming time of the model group was shortened (P<0.05). Compared with the model group, the exhaustive swimming time of mice in the high, medium and low dose groups of Guiqi Yiyuan ointment was increased (P<0.05). Compared with the blank group, the concentration of epinephrine and norepinephrine in the model group was decreased (P<0.01). Compared with the model group, the concentrations of adrenaline and norepinephrine in the high and medium dose groups of Guiqi Yiyuan ointment were significantly increased (P<0.01). Compared with the blank group, the levels of B cells and T lymphocyte subsets CD4+ and CD8+ in the model group were significantly decreased(P<0.01); compared with the model group, the levels of T lymphocyte subsets CD4+ and CD8+ in the high dose group of Guiqi Yiyuan ointment were significantly increased; compared with the blank group, the content of liver glycogen, muscle glycogen, spleen index and thymus index in the model group were significantly decreased (P<0.01). Compared with the model group, the contents of liver glycogen, muscle glycogen and spleen index in the high, medium and low dose groups of Guiqi Yiyuan ointment were significantly increased (P<0.01). Compared with the model group, the thymus index of high, medium and low dose groups of Guiqi Yiyuan ointment increased (P<0.05).Conclusion Guiqi Yiyuan ointment can significantly improve the transportation and immune function of mice with Qi deficiency induced by exhaustive swimming, and the effect of high and medium dose groups is more obvious.
