目的 基于癌症和肿瘤基因图谱(TCGA)数据库分析SERPINH1基因在肺腺癌中的表达及其与预后的关系。方法 获取TCGA数据库肺腺癌(TCGA-LUAD)RNA测序数据和患者临床资料,分析肺腺癌和正常肺组织中SERPINH1基因mRNA表达差异;分析SERPINH1表达水平与患者年龄、性别、临床分期、驱动基因状态、吸烟史、疾病进展和生存状态等临床特征的关系;以无进展间期(DFI)和总生存期(OS)为临床指标,绘制Kaplan-Meier生存曲线,建立多因素Cox比例风险模型,分析SERPINH1表达对肺腺癌生存的影响,并绘制预测模型的列线图。结果 SERPINH1在肺癌组织高表达(P<0.001);SERPINH1在死亡组表达量高于生存组(P<0.01)。ERPINH1高表达组年龄小于65岁患者比例更高,且女性患者比例高于男性(P<0.05);SERPINH1基因表达水平对患者DFI和OS均有影响(P<0.05),高表达提示肺腺癌患者DFI和OS更差。多变量Cox回归分析显示SERPINH1基因高表达是肺腺癌患者预后总生存差的独立风险因子(HR=1.32,95%CI:1.07~1.63,P=0.009)。结论 SERPINH1基因高表达是肺腺癌患者预后总生存差的独立风险因子,可有效预测肺腺癌患者预后。
Abstract:
Objective To analyze the expression of SERPINH1 gene in lung adenocarcinoma and its relationship with prognosis based on The Cancer Genome Atlas (TCGA) database. Methods The RNA sequencing data of lung adenocarcinoma in TCGA database (TCGA-LUAD) and the clinical data of patients were obtained, and the difference of SERPINH1 gene mRNA expression between lung adenocarcinoma and normal lung tissue was analyzed. The relationship between SERPINH1 expression level and clinical characteristics such as age, gender, clinical stage, driver gene status, smoking history, disease progression and survival status was analyzed. Kaplan-Meier survival curve was drawn with progression-free interval (DFI) and overall survival (OS) as clinical indicators. Multivariate Cox proportional hazard model was established to analyze the effect of SERPINH1 expression on the survival of lung adenocarcinoma, and the nomogram of the prediction model was drawn. Results SERPINH1 was highly expressed in lung cancer tissues (P<0.001). The expression of SERPINH1 in the death group was higher than that in the survival group (P<0.01). The proportion of patients younger than 65 years old in the ERPINH1 high expression group was higher, and the proportion of female patients was higher than that of male patients (P<0.05). The expression level of SERPINH1 gene had an effect on DFI and OS of patients (P<0.05), and high expression suggested that DFI and OS of patients with lung adenocarcinoma were worse. Multivariate Cox regression analysis showed that high expression of SERPINH1 gene was an independent risk factor for poor overall survival in patients with lung adenocarcinoma (HR=1.32, 95%CI: 1.07-1.63, P=0.009). Conclusion High expression of SERPINH1 gene is an independent risk factor for poor overall survival in patients with lung adenocarcinoma, which can effectively predict the prognosis of patients with lung adenocarcinoma.
