摘要:
目的 初步探索神经生长因子受体在喉癌Hep2细胞系的表达情况并研究相关的细胞生物学特性。方法 应用免疫细胞化学及免疫荧光染色法,检测p75NTR在喉癌Hep2细胞系中的表达;应用流式细胞仪检测人喉癌Hep2细胞系中呈p75NTR高表达及CD133高表达细胞的分布比例情况。结果 喉癌Hep2细胞系内小部分肿瘤细胞显示免疫染色强阳性,p75NTR强阳性显色的细胞部分处于旺盛的增殖分裂状态;大部分呈CD133强阳性的Hep2细胞也呈p75NTR强阳性。结论 p75NTR高表达于喉癌Hep2细胞系中具有较强增殖活力的细胞,p75NTR信号通路对支持或促进喉癌细胞增殖具有潜在作用。p75NTR高表达的喉癌Hep2细胞与肿瘤干细胞之间的关系密切,有待进一步深入研究。
Abstract:
Abstract:Objective To investigate the expression of nerve growth factor receptor in laryngeal squamous cell carcinoma Hep2 cell line and to study the related cellular biological characteristics.Methods Immunocytochemistry and immunofluorescence staining were used to detect the expression of p75NTR in Hep2 cell line of laryngeal squamous cell carcinoma,and the distribution of p75NTR and CD133 in Hep2 cell line of laryngeal squamous cell carcinoma was detected by flow cytometry.Results A small number of tumor cells in the Hep2 cell line of laryngeal squamous cell carcinoma showed strong immunostaining,and the cells with strong positive coloration of p75NTR were in a proliferative and dividing state;most of the Hep2 cells positive for CD133 were also strongly positive for p75NTR.Conclusion p75NTR is highly expressed in the Hep2 cell line of laryngeal squamous cell carcinoma and has strong proliferative activity.The p75NTR signaling pathway has a potential role in supporting or promoting the proliferation of laryngeal squamous cell carcinoma cells.The relationship between p75NTR-expressing laryngeal squamous cell carcinoma Hep2 cells and tumor stem cells is closely related and needs further study.
