目的 研究E盒结合锌指蛋白1和叉头框家族蛋白在非小细胞肺癌组织中的表达,并探讨其表达对非小细胞肺癌浸润、转移的影响。方法 收集2013年6月~2015年6月沧州市中心医院手术切除的130组非小细胞肺癌肿瘤组织和癌旁正常组织,免疫组织化学法和免疫印迹法检测E盒结合锌指蛋白1和叉头框家族蛋白蛋白的表达,分析E盒结合锌指蛋白1和叉头框家族蛋白与非小细胞肺癌临床资料的关系。结果 肿瘤组织E盒结合锌指蛋白1蛋白表达显著高于癌旁正常组织(P<0.05),而叉头框家族蛋白蛋白表达显著低于癌旁正常组织(P<0.05);E盒结合锌指蛋白1和叉头框家族蛋白蛋白表达均与肿瘤大小、分化程度、淋巴结转移、远端转移以及TNM分期显著相关(P<0.05);在淋巴结转移或远端转移的非小细胞肺癌患者中,E盒结合锌指蛋白1蛋白表达显著升高(P<0.05),叉头框家族蛋白蛋白表达显著降低(P<0.05);高表达E盒结合锌指蛋白1或者低表达叉头框家族蛋白的非小细胞肺癌患者术后2年生存率显著降低。结论 E盒结合锌指蛋白1在非小细胞肺癌组织中高表达,叉头框家族蛋白在非小细胞肺癌组织中低表达,两者协同促进非小细胞肺癌浸润转移。
Abstract:
Objective To study the expression of zinc finger E binding protein 1 and forkhead box family proteins in non small cell lung cancer,and to investigate the expression of non small cell lung cancer, metastasis.Methods From June 2013 to June 2015,130 groups of non-small cell lung cancer(NSCLC)tumor tissues and adjacent normal tissues were collected from Cangzhou central hospital. Immunohistochemistry and western blotting were used to detect the expression of E-box binding zinc finger protein 1 and fork frame family protein.To analyze the relationship between E-box binding zinc finger protein 1 and fork box family protein and clinical data of non-small cell lung cancer.Results The expression of E-box binding zinc finger protein-1 protein in tumor tissues was significantly higher than that in adjacent normal tissues(P<0.05),while the protein expression of fork box family protein was significantly lower than that in adjacent normal tissues(P<0.05).The expression of E-box binding zinc finger protein 1 and fork box family protein were significantly correlated with tumor size,differentiation,lymph node metastasis,distal metastasis and TNM stage(P<0.05).In non-small cell lung cancer patients with lymph node metastasis or distant metastasis,the expression of E-box combined with zinc finger protein 1 (P<0.05)and protein of fork-framed family protein decreased significantly(P<0.05).High expression patients with non-small-cell lung cancer with E-box combined with zinc finger protein 1 or with low expression of fork-frame family protein had a significantly lower 2-year postoperative survival rate.Conclusion The E-box combined with zinc finger protein 1 is highly expressed in non-small cell lung cancer tissues,and the forkhead box family protein is low expressed in non-small cell lung cancer tissues.Both of them can promote the invasion and metastasis of non-small cell lung cancer.
