摘要:
目的 探讨LMP1羧基端功能活性区促进鼻咽癌干细胞SP18增殖作用机制。方法 采用生长曲线、平皿克隆形成实验与软琼脂集落实验分析LMP1TRADD对SP18细胞增殖的作用;运用Western-blot检测SP18细胞中Wnt1、β-catenin与p-β-catenin蛋白的表达。结果 SP18-LMP1TRADD细胞的增殖能力较SP18-LMP1细胞减弱(P<0.01);与SP18-LMP1细胞比较,SP18-LMP1TRADD细胞中Wnt1与β-catenin表达降低,而p-β-catenin蛋白表达增加(P<0.01)。结论 LMP1经Wnt1/β-catenin途径促进鼻咽癌干细胞SP18的增殖。
Abstract:
Abstract:Objective To investigate the mechanism of the carboxyl-terminal functional active region of LMP1 in promoting proliferation of nasopharyngeal carcinoma SP18 stem cells.Methods Growth curve,plate colony formation assay and soft agar colony assay were used to analyze the effect of LMP1TRADD on the proliferation of SP18 cells.The expression of Wnt1,β -catenin and p-β-catenin in SP18 cells was detected by Western-blot.Results The proliferative ability of SP18-LMP1TRADD cells was lower than that of SP18-LMP1 cells(P<0.01),compared with SP18-LMP1 cells, the expression of Wnt1 and β-catenin in SP18-LMP1TRADD cells decreased,while the expression of p-β-catenin protein increased(P<0.01).Conclusion LMP1 can promote the proliferation of SP18 in nasopharyngeal carcinoma stem cells via Wnt1/β-catenin pathway.
