摘要:
目的 建立一种高效液相色谱-质谱联用技术(LC-MS/MS)测定人血浆盐酸伪麻黄碱浓度的方法。方法 血浆样品经液液萃取后,以甲醇∶水∶甲酸(60∶40∶1,V/V/V)为流动相,流速0.35 ml/min,采用Cap cell pack C18色谱柱分离,150 mm×2.1 mm, 5 μm(USA);柱温20℃,选择离子喷雾离子化源串联质谱,通过正离子方式检测;选择反应监测(MRM)方式转化m/z=166.3/148.2(盐酸伪麻黄碱)和m/z= 152.2/134.15(内标苯丙醇胺)进行定量分析,用内标法测定含量。结果 伪麻黄碱浓度分别在5.0~400.0 ng/ml的范围内线性良好,R=0.9962,最低检测限为1.0 g/ml。结论 本方法操作简便、快速,结果准确,可用于该药物的含量测定,同时也可为临床药动学研究提供一定的参考。
Abstract:
Objective To establish a high performance liquid chromatography-mass spectrometry (LC-MS/MS) method for the determination of pseudoephedrine hydrochloride in human plasma.Methods After the liquid sample was extracted by liquid-liquid extraction, the mobile phase was methanol:water:formic acid (60∶40∶1, V/V/V), the flow rate was 0.35 ml/min, and separated by Cap cell pack C18 column, 150 mm×2.1mm,5 μm (USA); column temperature 20°C, selective ion spray ionization source tandem mass spectrometry, positive ion detection; selective reaction monitoring (MRM) conversion m/z=166.3/148.2 (pseudoephedrine hydrochloride) and m/z=152.2/134.15 (internal standard phenylpropanolamine) for quantitative analysis, content determined by internal standard method.Results The pseudoephedrine concentration was linear in the range of 5.0~400.0 ng/ml,R=0.9962, and the minimum detection limit was 1.0 g/ml.Conclusion The method is simple, rapid and accurate, and can be used for the determination of the drug content. It can also provide a reference for clinical pharmacokinetic studies.
