摘要:
目的 使用UPLC测定健脾益肺口服液中白术内酯Ⅰ、白术内酯Ⅲ的含量。方法 采用Waters H-class 超高效液相色谱仪测定白术内酯Ⅰ、白术内酯Ⅲ的含量,色谱柱:Waters BEH C18(50 mm×2.1 mm,1.7 μm),柱温:30℃,进样量:2 μl,检测波长:220 nm,流动相:乙腈(A)-水(B),梯度洗脱,流速:0.2 ml/min。结果 白术内酯Ⅰ线性方程为y=0.0528x+0.0241(r=0.9998),线性范围2~160 μg/ml;白术内酯Ⅲ的线性方程为y=0.0786x(r=0.9998),线性范围3~140 μg/ml;白术内酯Ⅰ及白术内酯Ⅲ的精密度、重复性、稳定性、加样回收率均符合要求。结论 初步建立了健脾益肺口服液中白术内酯Ⅰ、白术内酯Ⅲ含量测定的高效、迅速的检测方法,为下一步质量标准的提高奠定基础。
Abstract:
Objective To determine the content of atractylenolide I and atractylenolide III in Jianpi Yifei Oral Liquid by UPLC. Methods The contents of atractylenolide I and atractylenolide III were determined by Waters H-class ultra performance liquid chromatography. Column: Waters BEH C18 (50 mm × 2.1 mm, 1.7 μm), column temperature: 30°C, injection Amount: 2 μl, detection wavelength: 220 nm, mobile phase: acetonitrile (A)-water (B), gradient elution, flow rate: 0.2 ml/min. Results The linear equation of atractylenolide I was y=0.0528x+0.0241 (r=0.9998), the linear range was 2~160 μg/ml; the linear equation of atractyl lactone III was y=0.0786x (r=0.9998), linear range 3 ~140 μg/ml; the precision, repeatability, stability and recovery rate of atractylenolide I and atractylenolide III meet the requirements. Conclusion The high-efficiency and rapid detection method for the determination of atractylenolide I and atractylenolide III in Jianpi Yifei Oral Liquid was established, which laid a foundation for the improvement of quality standards.
