摘要:
目的 观察肺腺癌组织中miRNA-21的表达情况,为miRNA-21成为新的肺腺癌靶点基因提供临床依据。方法 收集自2017年6月~2019年6月广西医科大学第二附属医院共53例肺腺癌患者的癌组织、配对癌旁组织以及12例非癌疾病肺组织。采用实时荧光定量PCR法(qPCR)检测其miRNA-21和PTEN的表达情况,并分析二者的相关性。结果 肺腺癌组织中miRNA-21表达高于癌旁组织和非癌肺组织,其中Ⅲ~Ⅳ期患者中高于Ⅰ~Ⅱ期患者;肺腺癌组织中PTEN低于癌旁组织和非癌肺组织,其中Ⅲ~Ⅳ期患者中低于Ⅰ~Ⅱ期患者,差异有统计学意义(P<0.05); miRNA-21和PTEN的表达呈负相关(r=-0.663,P<0.05)。结论 miRNA-21在肺腺癌组织中抑制了PTEN的表达,miRNA-21极有可能成为肺腺癌的治疗靶点。
Abstract:
Objective To observe the expression of miRNA-21 in lung adenocarcinoma tissues and provide a clinical basis for miRNA-21 to become a new target gene for lung adenocarcinoma.Methods From June 2017 to June 2019, 53 cases of lung adenocarcinoma patients’ cancer tissues, paired paracancerous tissues and 12 cases of non-cancer disease lung tissues were collected from the Second Affiliated Hospital of Guangxi Medical University. Real-time fluorescence quantitative PCR (qPCR) was used to detect the expression of miRNA-21 and PTEN, and to analyze the correlation between the two.Results The expression of miRNA-21 in lung adenocarcinoma tissues was higher than that in paracancerous tissues and non-cancerous lung tissues. Among them, stage III to IV patients were higher than those in stage I to II patients; PTEN in lung adenocarcinoma tissues was lower than that in paracancerous tissues and noncancerous lung tissues.Among them, the patients in stage III to IV were lower than those in stage I to II, the difference was statistically significant (P<0.05); the expression of miRNA-21 and PTEN was negatively correlated(r=-0.663,P<0.05).Conclusion miRNA-21 inhibits the expression of PTEN in lung adenocarcinoma tissues. miRNA-21 is very likely to be a therapeutic target for lung adenocarcinoma.
