目的 探讨温度对脐带血中单个核细胞(MNC)和干细胞的影响。方法 采集2020年7月~10月我院16例产妇的脐带血,均取30 ml,平均分为4 ℃组和室温组(RT组),均放置12 h,应用密度梯度离心法分离单个核细胞,用血细胞分析仪检测MNC数,流式细胞术检测干细胞(CD34+细胞)占CD45+细胞的比例,计算CD34+细胞数;比较两组MNC数、CD34+细胞比例及CD34+细胞数,分析不同温度下CD34+细胞数与MNC的相关性。结果 4 ℃组与室温组MNC数分别为(9.28±4.65)×108/L、(20.11±8.37)×108/L,组间差异有统计学意义(P<0.05);4 ℃组与室温组CD34+细胞比例分别为(1.10±0.56)%、(0.55±0.24)%,组间差异有统计学意义(P<0.05);4 ℃组与室温组CD34+细胞数分别为(9.41±5.08)×106/L、(10.80±5.40)×106/L,差异无统计学意义(P>0.05);4 ℃组CD34+细胞数与MNC无相关性(r=0.295,P=0.267),室温组CD34+细胞数与MNC呈正相关(r=0.551,P=0.027)。结论 室温保存脐带血可有效保护干细胞和MNC,4 ℃保存可保护干细胞但损失MNC。
Abstract:
Objective To investigate the impact of temperature on mononuclear cell (MNC) and stem cell in umbilical cord blood (UCB).Methods Sixteen umbilical cord blood units were collected in our hospital between July 2020 and October 2020. Thirty milliliters UCB were taken and then divided into two groups including 4℃group and room temperature (RT) group. They were kept at 4℃ or RT for 12 hours. MNCs were separated using Ficoll-Hypaque density centrifugation and MNC number was measured using automated hematology analyzer. CD34+ cell ratio was analyzed using flow cytometry and CD34+ cell number was further calculated. MNC number, CD34+ cell ratio and CD34+ cell number were compared between 4 ℃ group and RT group. Correlations between CD34+ cell number and MNC number were analyzed in each group.Results MNC number was lower in 4℃ group than RT group [(9.28±4.65)×108/L vs (20.11±8.37)×108/L, P<0.001]. CD34+ cell ratio was higher in 4 ℃ group than RT group [(1.10±0.56)% vs (0.55±0.24)%, P=0.001]. CD34+ cell number was not significantly different between 4 ℃ group and RT group[ (9.41±5.08)×106/L vs (10.80±5.40)×106/L, P=0.225]. CD34+ cell number was not correlated with MNC number in 4℃ group (r=0.295, P=0.267) while they were positively correlated in RT group (r =0.551, P=0.027).Conclusion Room temperature storage of UCB is useful for both stem cell and MNC while 4℃ storage is useful for stem cell only.
