摘要:
目的 探讨强力抗氧化剂丹酚酸B对睾丸缺血再灌注损伤的效应和相关机制。方法 于2021年1月-6月选择60只雄性SD大鼠,将其随机分为对照组、缺血再灌注组和治疗组,每组20只。对照组以1根11/0无创伤丝线穿过睾丸白膜,然后将睾丸放回阴囊,缝合皮肤切口;缺血再灌注组逆时针扭转左侧睾丸720°诱导睾丸缺血,然后用11/0丝线固定扭转睾丸到阴囊上,2 h后使睾丸恢复到正常位置;治疗组处理同缺血再灌注组,在再灌注时从尾静脉注射丹酚酸B治疗。比较各组睾丸中丙二醛含量、环磷酸腺苷反应元件调控因子τ(CREMτ)蛋白表达和生精功能。结果 缺血再灌注组同侧睾丸的丙二醛含量高于对照组和治疗组,CREMτ表达和生精功能低于对照组和治疗组,差异有统计学意义(P<0.05);丹酚酸B治疗明显降低了同侧睾丸的丙二醛含量,提高了CREMτ表达和生精功能(P<0.05)。三组间对侧睾丸的丙二醛水平、CREMτ表达和生精功能比较,差异无统计学意义(P>0.05)。对照组睾丸可观察到正常的曲细精管直径和生精细胞层数,在曲细精管内有许多成熟的精子和一个开放的管腔;缺血再灌注组同侧睾丸表现出曲细精管萎缩和生精细胞层数减少;治疗组同侧睾丸曲细精管的结构几乎符合正常标准,曲细精管内可见成熟的精子,但曲细精管的管腔内包含有脱落的生精细胞。结论 丹酚酸B可通过清除活性氧上调CREMτ的表达,其对睾丸缺血再灌注损伤有保护作用。
Abstract:
Objective To investigate the effect and related mechanism of strong antioxidant salvianolic acid B on testicular ischemia-reperfusion injury.Methods From January to June 2021, 60 male SD rats were selected and randomly divided into control group, ischemia-reperfusion group and treatment group, with 20 rats in each group. In the control group, an 11/0 non-invasive silk thread passed through the albuginea of the testis, then the testis was put back into the scrotum, and the skin incision was sutured. In the ischemia-reperfusion group, 720° of the left testis was twisted connterclock-wise to induce testicular ischemia, and then 11/0 silk thread was used to fix twisted testis to the scrotum. After 2 h, the testis was restored to normal position. The treatment group was treated the same as the ischemia-reperfusion group. Salvianolic acid B was injected from the tail vein during reperfusion. The malondialdehyde content, expression of cAMP response element modulator τ (CREMτ)protein and spermatogenic function in testis of each group were compared.Results The content of MDA in ipsilateral testis of ischemia-reperfusion group was higher than that of control group and treatment group, and the expression of CREMτ and spermatogenic function were lower than those of control group and treatment group, the difference was statistically significant (P<0.05), and Salvianolic acid B treatment significantly decreased MDA content in ipsilateral testis, increased CREMτ expression and spermatogenic function (P<0.05). There was no significant difference in MDA level, CREMτ expression and spermatogenic function in contralateral testes among the three groups (P>0.05). The normal diameter of seminiferous tubules and layers of spermatogenic cells were observed in the control group, and there were many mature sperms and an open lumen in the seminiferous tubules; testis showed atrophy of seminiferous tubules and reduction of spermatogenic cell layer in ipsilateral testes of ischemia reperfusion group; the structure of testicular seminiferous tubules in ipsilateral testes of the treatment group almost met the normal standard, and mature sperm could be seen in the seminiferous tubules, but the lumen of the seminiferous tubules contained exfoliated spermatogenic cells.Conclusion Salvianolic acid B can upregulate the expression of CREMτ by scavenging reactive oxygen species, which has a protective effect on testicular ischemia-reperfusion injury.
