目的 基于中国脑胶质瘤基因组图谱(CGGA)数据库,分析研究精氨酸甲基转移酶1(CARM1)在胶质瘤中的差异性表达情况,探讨其潜在的作用机制,进而为胶质瘤的诊断与治疗提供新的靶点。方法 基于CGGA数据库中胶质瘤患者的相关临床资料,分析不同情况胶质瘤患者中CARM1表达差异性及与患者生存率及生存时间的关系,最后通过String数据库对CARM1蛋白表达相关作用网络进行分析。结果 不同性别的胶质瘤患者中CARM1表达比较,差异无统计学意义(P=0.36);CGGA数据库分析显示,CARM1在原发性胶质瘤和复发性胶质瘤组织中呈高水平表达(P=1.4e-18);CARM1的表达与年龄有一定相关性(P=0.0006),与胶质瘤发生发展呈正相关(P=1e-16);CARM1在复发性胶质瘤组织中表达量高于原发性胶质瘤(P=0.0011);CARM1在原发性、复发性胶质瘤组织中的表达与患者生存期呈负相关(P<0.0001);利用String数据库分析显示,CARM1与EP300、TP53、NCOA3等多个蛋白有相互作用。结论 CARM1在胶质瘤组织中呈高表达,且与患者的预后密切相关,可作为诊断和治疗胶质瘤的潜在靶点。
Abstract:
Objective To analyze the differential expression of coactivator associated arginine methyltransferase 1 (CARM1) in glioma based on the Chinese Glioma Genome Atlas (CGGA) database, and to explore its potential mechanism, so as to provide new targets for the diagnosis and treatment of glioma.Methods The clinical data of glioma patients in CGGA database were used to analyze the difference of CARM1 expression in glioma patients under different conditions and the relationship between CARM1 expression and survival rate and survival time of patients. Finally, the correlation network of CARM1 expression protein was analyzed through String database.Results There was no significant difference in CARM1 between different genders (P=0.36). CGGA database analysis showed that CARM1 was highly expressed in primary glioma and recurrent glioma tissues (P=1.4e-18). The expression of CARM1 was correlated with age (P=0.0006), and positively correlated with the occurrence and development of glioma (P=1e-16). The expression of CARM1 in recurrent glioma was higher than that in primary glioma (P=0.0011). The expression of CARM1 in primary and recurrent glioma tissues was negatively correlated with the survival time of patients (P<0.0001). String database analysis showed that CARM1 interacted with EP300, TP53, NCOA3 and other proteins.Conclusion CARM1 is highly expressed in glioma tissues and is closely related to the prognosis of patients, so it can be used as a potential target for diagnosis and treatment of glioma.
