摘要:
目的 探讨黄芩苷镁对脂多糖(LPS)联合D-氨基半乳糖(D-GalN)致急性肝损伤大鼠炎症因子的影响。方法 取SPF级雄性SD大鼠54只,适应性饲养后随机分为正常组、模型组、黄芩苷镁高、中、低剂量组及异甘草酸镁组,各9只;尾静脉注射给药、造模,黄芩苷镁高、中、低剂量组分别注射100、50、25 mg/kg的黄芩苷镁,异甘草酸镁组尾静脉注射18 mg/kg异甘草酸镁,模型及正常组尾静脉注射生理盐水,比较各组IL-1β、IL-6、TNF-α及IFN-γ水平。结果 模型组IL-6、IL-1β、TNF-α、IFN-γ水平高于正常组,差异有统计学意义(P<0.05);黄芩苷镁高、中剂量组及异甘草酸镁组IL-6、IL-1β、IFN-γ水平低于模型组,差异有统计学意义(P<0.05);黄芩苷镁低剂量组IFN-γ水平低于模型组,差异有统计学意义(P<0.05);黄芩苷镁中、低剂量组IL-6、IL-1β、IFN-γ水平高于异甘草酸镁组,差异有统计学意义(P<0.05)。结论 黄芩苷镁预处理对于LPS联合D-GalN诱导的大鼠急性肝损伤具有保护作用,其作用可能与抑制炎性细胞因子产生有关。
Abstract:
Objective To investigate the effect of magnesium baicalin on inflammatory factors in rats with acute liver injury induced by lipopolysaccharide (LPS) combined with D-galactosamine (D-GalN).Methods A total of 54 SPF male SD rats were randomly divided into normal group, model group, the high, medium and low dose of baicalin magnesium groups and magnesium isoglycyrrhizinate group, with 9 rats in each group. The high, medium and low dose groups of baicalin magnesium were injected with 100, 50 and 25 mg/kg baicalin magnesium respectively. The magnesium isoglycyrrhizinate group was injected with 18 mg/kg magnesium isoglycyrrhizinate. The model and normal group were injected with normal saline. The levels of IL-1β, IL-6, TNF-α and IFN-γ in each group were compared.Results The levels of IL-6, IL-1β, TNF-α and IFN-γ in the model group were higher than those in the normal group (P<0.05). The levels of IL-6, IL-1β and IFN-γ in the high, medium dose of baicalin magnesium groups and magnesium isoglycyrrhizinate group were lower than those in model group, and the differences were statistically significant (P<0.05). The level of IFN-γ in the low dose of baicalin magnesium group was lower than that in the model group, and the difference was statistically significant (P<0.05). The levels of IL-6, IL-1β and IFN-γ in the medium and low dose of baicalin magnesium groups were higher than those in magnesium isoglycyrrhizinate group, and the differences were statistically significant (P<0.05).Conclusion Baicalin magnesium pretreatment has a protective effect on acute liver injury induced by LPS combined with D-GalN in rats, which may be related to the inhibition of inflammatory cytokines.
