摘要:
目的 探究RCN1在原发性肝癌患者中的表达水平,分析其与患者临床病理特征及预后的关系,探究其可能的生物功能。方法 在TCGA及UALCAN数据库分析RCN1在肝癌中的表达情况,采用R语言对TCGA Liver hepatocellular carcinoma(LIHC)数据进行可视化分析,明确RCN1表达与肝癌患者临床分期分级的关系,运用ROC曲线和Kaplan-Meier的方法评价RCN1对肝癌患者诊断和预后的评估价值,通过LinkedOmics平台对TCGA中肝癌患者的RCN1差异表达和共表达基因进行分析,取重叠基因进行GO和KEGG分析富集情况。结果 TCGA数据库显示,RCN1在肝癌患者临床样本中高表达(P<0.01)。RCN1的表达与患者病理TNM分期、分级、生存和预后相关联(P<0.05),RCN1在差异基因中呈上调,GO和KEGG分析显示RCN1基因主要参与核受体的活化,转录因子的激活等。通过PPI网络分析,找到ESR1等中心靶点。通过相关性分析,RCN1与ESR1相互作用,可能参与调控内质网应激,诱导肝癌细胞的恶性进展。结论 RCN1在肝癌中呈高表达,其表达与肝癌TNM分期和生存预后有关联,RCN1可能与核受体和转录因子的激活相关,与ESR1等基因共同调控肝癌细胞的恶性行为学。
Abstract:
Objective To investigate the expression level of RCN1 in patients with primary hepatocellular carcinoma (HCC) and analyze its relationship with the clinicopathological features and prognosis, and to explore its possible biological functions. Methods The expression of RCN1 in liver cancer was analyzed in TCGA and UALCAN databases. TCGA Liver hepatocellular carcinoma (LIHC) data were visualized by R language to clarify the relationship between RCN1 expression and clinical stage and grade of liver cancer patients. ROC curve and Kaplan-Meier method were used to evaluate the value of RCN1 in the diagnosis and prognosis of liver cancer patients. The differentially expressed and co-expressed genes of RCN1 in patients with liver cancer in TCGA were analyzed by LinkedOmics platform, and the overlapping genes were enriched by GO and KEGG analysis. Results TCGA database showed that RCN1 was highly expressed in clinical samples of HCC patients (P<0.01). The expression of RCN1 was associated with pathological TNM stage, grade, survival and prognosis of patients (P<0.05). RCN1 was up-regulated in differential genes. GO and KEGG analysis showed that RCN1 gene was mainly involved in the activation of nuclear receptors and the activation of transcription factors. Through PPI network analysis, central targets such as ESR1 were found. And through correlation analysis, the interaction between RCN1 and ESR1 might be involved in the regulation of endoplasmic reticulum stress and induce the malignant progression of liver cancer cells. Conclusion RCN1 is highly expressed in hepatocellular carcinoma, and its expression is associated with TNM stage and survival prognosis of hepatocellular carcinoma. While, RCN1 may be related to the activation of nuclear receptors and transcription factors, and regulate the malignant behavior of hepatocellular carcinoma cells together with ESR1 and other genes.
