摘要:
目的 探讨miR-200c对颅咽管瘤侵袭性的影响,并阐述其作用机制。方法 选择造釉细胞型及鳞状上皮乳头型颅咽管瘤原代细胞为对象,研究miR-200c在原代细胞中的表达情况。通过对造釉细胞型颅咽管瘤原代细胞过表达和敲低miR-200c,研究miR-200c对造釉细胞型颅咽管瘤侵袭性的影响及作用机制。结果 与鳞状上皮乳头型颅咽管瘤原代细胞组相比,造釉细胞型颅咽管瘤原代细胞组中miRNA-200c的表达下降,统计学意义极显著(P<0.001);与对照组相比,miR-200c mimics组结晶紫染色细胞数量减少(P<0.05),而miR-200c inhibitor组结晶紫染色细胞数量增加(P<0.05),miR-200c inhibitor组细胞数多于miR-200c mimics组,统计学意义显著(P<0.01)。结论 miRNA-200c在造釉细胞型颅咽管瘤原代细胞中表达明显减少,促进了EMT的发生,导致造釉细胞型颅咽管瘤的侵袭性增加。
Abstract:
Objective To explore the effect of miR-200c on the invasiveness of craniopharyngioma and clarify its mechanism of action.Methods The primary cells of adamantinomatous craniopharyngiomas and papillary craniopharyngiomas were selected to study the expression of miR-200c in primary cells. Through overexpression and knockdown of miR-200c in primary cells of adamantinomatous craniopharyngiomas, the effect of miR-200c on the invasiveness of adamantinomatous craniopharyngiomas and its mechanism were studied.Results Compared with the primary cell group of squamous papillary craniopharyngioma, the expression of miRNA-200c in the primary cell group of adamantinomatous craniopharyngioma decreased significantly (P<0.001). Compared with the control group, the number of crystal violet staining cells in the miR-200c mimics group decreased (P<0.05), while the number of crystal violet staining cells in the miR-200c inhibitor group increased (P<0.05), and the number of cells in the miR-200c inhibitor group was more than that in the miR-200c mimics group (P<0.01).Conclusion The expression of miRNA-200c in primary cells of adamantinomatous craniopharyngioma is significantly reduced, which promotes the occurrence of EMT and increased the invasiveness of adamantinomatous craniopharyngioma.
