摘要:
目的 研究蹄叶橐吾醇化乙酸乙酯萃取物(EEOLF)对金黄色葡萄球菌的抗菌消炎作用。方法 测定EEOLF对金黄色葡萄球菌的最小抑菌浓度(MIC);取正常培养的巨噬细胞RAW264.7为空白组,采用金黄色葡萄球菌作用于RAW264.7细胞建立体外实验模型(模型组)。细胞活/死染色观察EEOLF(剂量为5 μg/ml)作用于模型组后细胞的损伤情况,ELISA法测定EEOLF(剂量为1.25、2.5 及5 μg/ml)作用于模型组后IL-1β及TNF-α的释放量。结果 EEOLF对金黄色葡萄球菌生长有抑制作用,最小抑菌浓度为1300 μg/ml;当金黄色葡萄球菌作用于RAW264.7细胞后,细胞发生严重损伤,同时IL-1β及TNF-α的分泌量增加(P<0.05)。对金黄色葡萄球菌感染的细胞给药后,药物浓度为1.25 μg/ml和2.5 μg/ml时,炎症因子的分泌量没有明显减少(P>0.05),当药物浓度为5 μg/ml时,炎性因子L-1β及TNF-α的分泌量下降(P<0.05),同时细胞损伤情况改善。结论 EEOLF能够抑制金黄色葡萄球菌生长,也能缓解金黄色葡萄球菌引起的炎症反应,其抗菌消炎作用值得深入研究。
Abstract:
bjective To study the antibacterial and anti-inflammatory effects of ethanolic extract of ligulariafischeri (EEOLF) on Staphylococcus aureus.Methods The minimum inhibitory concentration (MIC) of EEOLF against Staphylococcus aureus was determined. Normal cultured macrophages RAW264.7 were taken as blank group, and Staphylococcus aureus was applied to RAW264.7 cells to establish cell inflammation model group in vitro (model group). The cell damage after EEOLF (5 μg/ml) in the model group was detected by living/dead staining, and the release of IL-1β and TNF-αafter EEOLF (1.25, 2.5 and 5 μg/ml) in the model group was determined by ELISA.Results EEOLF could inhibit the growth of Staphylococcus aureus, and the MIC was 1300 μg/ml. When Staphylococcus aureus acted on RAW264.7 cells, the cells were severely damaged, and the secretion of IL-1β and TNF-α was increased (P<0.05). When the drug concentration was 1.25 μg/ml and 2.5 μg/ml, the secretion of inflammatory factors was not significantly reduced (P>0.05). When the drug concentration was 5 μg/ml, the secretion of inflammatory factors L-1β and TNF-α was decreased (P<0.05), and the cell damage was improved.Conclusion EEOLF can inhibit the growth of Staphylococcus aureus and alleviate the inflammatory response caused by Staphylococcus aureus, and its antibacterial and anti-inflammatory effects are worthy for further study.
