目的 探究脯氨酸-谷氨酸-亮氨酸富集蛋白1(PELP1)在上皮性卵巢癌血管生成中的作用。方法 选取正常卵巢上皮细胞IOSE80及上皮性卵巢癌细胞OVCAR-3、Hey,设置Hey-Ctrl组为空白对照,Hey-OE-Vector组为阴性对照,Hey-OE-PELP1组为过表达组。实时荧光定量PCR(RT-qPCR)、Western Blot(WB)检测PELP1相对表达水平,WB检测PELP1相对表达水平,酶联免疫吸附实验(ELISA)测定OE-Vector-CM组、OE-PELP1-CM组上清液(CM)中VEGFA浓度。Transwell及血管生成法检测CM对人脐静脉内皮细胞(HUVECs)迁移及成管的影响。结果 上皮性卵巢癌细胞OVCAR-3、Hey中PELP1表达水平高于正常卵巢上皮细胞IOSE80细胞(P<0.05);Hey-OE-PELP1组PELP1表达水平高于Hey-Ctrl组及Hey-OE-Vector组(P<0.05);OE-PELP1-CM组VEGFA浓度、迁移细胞的数量、成管节点数量均高于OE-Vector-CM组(P<0.05)。结论 PELP1可促进上皮性卵巢癌的血管生成。
Abstract:
Objective To investigate the role of proline-glutamic acid-leucine-rich protein 1 (PELP1) in angiogenesis of epithelial ovarian cancer. Methods Normal ovarian epithelial cells IOSE80 and epithelial ovarian cancer cells OVCAR-3 and Hey were selected. Hey-Ctrl group was set as blank control, Hey-OE-Vector group as negative control, and Hey-OE-PELP1 group as overexpression group. The relative expression level of PELP1 was detected by real-time fluorescence quantitative PCR (RT-qPCR) and Western Blot (WB). The relative expression level of PELP1 was detected by WB. The concentration of VEGFA in the supernatant (CM) of OE-Vector-CM group and OE-PELP1-CM group was determined by enzyme-linked immunosorbent assay (ELISA). The effects of CM on the migration and tube formation of human umbilical vein endothelial cells (HUVECs) were detected by Transwell and angiogenesis methods. Results The expression level of PELP1 in epithelial ovarian cancer cells OVCAR-3 and Hey was higher than that in normal ovarian epithelial cells IOSE80 (P<0.05). The expression level of PELP1 in Hey-OE-PELP1 group was higher than that in Hey-Ctrl group and Hey-OE-Vector group (P<0.05). The concentration of VEGFA, the number of migrated cells and the number of tube nodes in OE-PELP1-CM group were higher than those in OE-Vector-CM group (P<0.05). Conclusion PELP1 promotes angiogenesis in epithelial ovarian cancer.
