摘要:
目的 观察甲氨蝶呤(MTX)对类风湿关节炎(RA)大鼠滑膜细胞凋亡及细胞凋亡调控基因的影响,探讨MTX的作用机制。方法 选择SPF级雌性健康SD大鼠40只,每只大鼠尾根部皮下多点注射Ⅱ型胶原蛋白0.2mg,造模后l0 d将动物随机分为假模型组、模型组,高(30mg/kg)、中(3mg/kg)、低(0.3mg/kg)给药组,各8只,每组按10 ml/kg MTX灌胃,假模型组、模型组给予等量生理盐水灌胃,1.0 g/kg。分离培养大鼠膝关节滑膜组织,采用流式细胞术检测细胞凋亡率、WB技术检测Caspase-3,Caspase-9,Bax,Bcl-2蛋白表达情况。结果 高、中、低剂量给药组抑制大鼠足肿胀的作用均优于模型组,差异有统计学意义(P<0.05),且呈剂量依赖性;各组均能不同程度诱导RA滑膜细胞凋亡,高剂量给药组与模型组及假模型组比较,诱导RA滑膜细胞凋亡更明显(P<0.05);高剂量给药组Caspase-3、Caspase-9、Bax蛋白表达量与模型组比较明显上调,Bc1-2蛋白表达量下降(P<0.05)。结论 MTX诱导风湿性关节炎大鼠滑膜细胞凋亡,其机制可能与下调Bcl-2表达,上调Caspase-3、Caspase-9与Bax表达有关。
Abstract:
Objective To observe the effects of methotrexate (MTX) on the apoptosis of synovial cells and apoptosis-regulating genes in rats with rheumatoid arthritis (RA), and to explore the mechanism of action of MTX. Methods 40 SPF female healthy SD rats were selected. Each rat was injected subcutaneously with 0.2mg of type Ⅱ collagen at the subcutaneous point.On the 10th d after modeling, the animals were randomly divided into a sham model group, a model group, a high (30 mg/kg), a medium (3 mg/kg), and a low (0.3 mg/kg) administration group.Each group consisted of 8 rats. Each group was administrated with 10 ml / kg MTX. The sham model group and model group were given the same amount of normal saline, 1.0 g/kg. The knee synovial tissue of rats was isolated and cultured, and the apoptosis rate was detected by flow cytometry, and the expression of Caspase-3, Caspase-9, Bax, Bcl-2 was detected by WB technology. Results The high, medium and low dose groups were better than the model group in inhibiting the swelling of the rats’ feet,the difference was statistically significant (P<0.05) and was dose-dependent; each group could induce RA synovial cells to varying degrees Apoptosis, compared with the model group and the pseudo-model group, the high-dose administration group induced apoptosis of RA synovial cells more significantly (P<0.05); the high-dose administration group expressed Caspase-3, Caspase-9, and Bax proteins with The model group was significantly up-regulated, and the expression of Bc1-2 protein decreased (P<0.05). Conclusion MTX induces apoptosis of synovial cells in rheumatoid arthritis rats. The mechanism may be related to down-regulating Bcl-2 expression, up-regulating Caspase-3, Caspase-9 and Bax expression.
