核转录因子κB(NFκB)是一类具有多向调节作用的转录调控因子,且在多个基因的转录及表达过程中发挥重要作用,与炎性相关疾病、肿瘤、疼痛等相关疾病相关。核转录因子抑制蛋白(IκB)主要由核转录因子κB抑制因子(NFκBIA)基因编码,参与调节NFκB信号通路,抑制NFκB功能,其同样与多种炎症性疾病、自身免疫性疾病及肿瘤的易感性有关。近年来,NFκB及其抑制因子在炎症、免疫及肿瘤等方面的影响一直广受关注。此外,NFκB信号通路与疼痛的调节也关系密切,但具体作用机制尚需进一步研究证实。本文主要对NFκB和IκB的结构、分布及表达情况,以及NFκB及NFκBIA基因在炎性疾病、肿瘤和慢性疼痛等多种疾病中的研究进展进行综述,旨在提高临床对其的认识。
Abstract:
Nuclear factor kappa-B (NFκB) is a multifaceted transcriptional regulator that plays an important role in the transcription and expression of multiple genes and is associated with inflammatory diseases, tumors, pain and other diseases.Nuclear transcription factor inhibitor protein (IκB), which is encoded by the Nuclear factor kappa-b inhibitor alpha (NFκBIA) gene, is involved in the regulation of the NFκB signaling pathway and the inhibition of NFκB function. It is also associated with a variety of inflammatory, autoimmune, and tumor susceptibility.The role of NFκB and its suppressor (NFκBIA) in inflammation, immunity, and tumor formation has received considerable attention in recent years.The NFκB signaling pathway is also closely related to pain regulation, but further studies are needed to confirm the specific mechanism.The structure, distribution and expression of NFκB and IκB were introduced and the research progress of NFκB and NFκBIA genes in inflammatory diseases, tumors and chronic pain are reviewed, aiming to improve clinical understanding of them.
