摘要:
目的 探讨倒置显微镜下观察狗肾上皮细胞(MDCK)分离流感病毒状态变化,为科学防控流感提供技术支持。方法 选取天津市津南区疾病预防控制中心流感网络实验室于2018年10月11日~20日流感病毒检测阳性的咽拭子样本20例,接种到MDCK传代细胞中,于接种后使用倒置显微镜观察细胞形态变化,包括细胞肿胀、间隙增大、核固缩或破裂、细胞部分或全部脱落,用血凝实验方法测定MDCK细胞不同分离状态下的流感病毒滴度状况。结果 倒置显微镜可以观察到MDCK细胞分离流感病毒状态的变化,阴性对照与未出现倒置显微镜镜下病变的MDCK细胞不能使人“O”型红细胞凝集;出现病变的MDCK细胞可以使人“O”型红细胞凝集,产生HA滴度。结论 倒置显微镜在观察MDCK细胞分离流感病毒过程中起到重要作用,可提前判断病毒的滴度与活力,为科学防控流感提供技术支持。
Abstract:
Abstract:Objective To investigate the changes of influenza virus status in dog kidney epithelial cells (MDCK) under inverted microscope and provide technical support for scientific prevention and control of influenza. Methods 20 cases of swallow swab samples with positive influenza virus detection were selected from the influenza network laboratory of Tianjin Jinnan District Center for Disease Control and Prevention from October 11 to 20, 2018. They were inoculated into MDCK passage cells and observed under inverted microscope after inoculation. Cell morphology changes, including cell swelling, gap enlargement, nuclear pyknosis or rupture, partial or total detachment of cells, blood clotting experiments were used to determine the influenza virus titer status of MDCK cells under different separation conditions. Results Inverted microscope can observe the changes of MDCK cells in the state of influenza virus isolation. Negative control and MDCK cells without inverted microscopic lesions cannot agglutinate human "O" type red blood cells; MDCK cells with lesions can make human "O" type Red blood cells aggregate to produce HA titers. Conclusion Inverted microscope plays an important role in the observation of MDCK cells in the isolation of influenza virus. It can judge the titer and vitality of virus in advance and provide technical support for scientific prevention and control of influenza.
