目的 探索GAS2L3在肝癌中的表达,探讨其潜在的机制。方法 通过qRT-PCR技术检测GAS2L3在肝正常组织和肝癌组织中的表达差异,用癌症基因组图谱(TCGA)数据库和基因表达综合数据库(GEO)进行验证,利用ROC曲线评价GAS2L3对肝癌诊断的准确性,采用Kaplan-Meier法、单因素及多因素COX回归分析GAS2L3表达与肝癌预后关系,利用GEO数据库验证生存关系,利用GAS2L3与临床变量构建列线图,基于GAS2L3差异分析获得基因功能途径富集,通过ESTIMATE、CIBERSORT及TIMER数据库分析GAS2L3与免疫浸润的相关性。结果 经过生信分析和实验验证发现,GAS2L3在肝癌组织中的表达量高于正常组织,并且其表达与肝癌的T分期、临床病理分期以及肝癌分级密切相关。GAS2L3对肝癌的诊断的AUC为0.827。同时,生存分析显示GAS2L3高的患者预后差;单因素和多因素COX分析显示,高表达的GAS2L3可作为肝癌的独立生物标志物。此外,GO、KEGG和GSEA富集分析显示,GAS2L3可能参与细胞核分裂和减数分裂细胞周期过程,主要富集于细胞周期、神经活性配体-受体相互作用和尼古丁成瘾通路。GAS2L3表达与肿瘤纯度和免疫浸润相关。结论 GAS2L3在肝癌中高表达,肝癌中GAS2L3的上调与低生存率和免疫浸润显著相关,GAS2L3可作为肝癌预后的潜在生物标志物和免疫治疗靶点。
Abstract:
Objective To explore the expression of GAS2L3 in liver cancer and its potential mechanism. Methods The expression difference of GAS2L3 in normal liver tissue and liver cancer tissue was detected by qRT-PCR, and verified by Cancer Genome Atlas (TCGA) database and integrated gene expression database (GEO). ROC curve was used to evaluate the accuracy of GAS2L3 in the diagnosis of liver cancer. Kaplan-Meier method, Univariate and multivariate COX regression analysis was performed to analyze the relationship between GAS2L3 expression and prognosis of liver cancer. GEO database was used to verify the survival relationship. A column graph was constructed using GAS2L3 and clinical variables. The correlation between GAS2L3 and immune infiltration was analyzed by using ESTIMATE, CIBERSORT and TIMER databases. Results After bioinformatics analysis and experimental verification, it was found that the expression of GAS2L3 in liver cancer tissues was higher than that of normal tissues, and its expression was closely related to the T-stage, clinical pathological stage and liver cancer grading of liver cancer. The AUC for the diagnosis of liver cancer by GAS2L3 was 0.827. At the same time, survival analysis showed that patients with high GAS2L3 had poor prognosis, and single- and multi-factor COX analysis showed that highly expressed GAS2L3 could serve as an independent biomarker for liver cancer. In addition, GO, KEGG and GSEA enrichment points suggested that GAS2L3 might be involved in nuclear division and meiotic cell cycle processes, mainly enriched in cell cycle, neuroactive ligand-receptor interactions and nicotine addiction pathways. GAS2L3 expression correlates with tumor purity and immune infiltration. Conclusion GAS2L3 is highly expressed in liver cancer, and GAS2L3 up-regulation in liver cancer is significantly associated with low survival rate and immune infiltration. GAS2L3 can be used as a potential biomarker and immunotherapy target for liver cancer prognosis.
