摘要:
目的 探讨滋阴潜阳养血安神法对室性心律失常大鼠心电图及对Nav1.5电流的影响。方法 将健康雄性SD大鼠30只随机分为模型组、对照组,滋阴潜阳养血安神组,每组10只。模型组给予生理盐水灌胃,对照组给予美托洛尔灌胃,滋阴潜阳养血安神组给予定悸复脉汤灌胃,灌胃2周后,通过生物机能实验系统BL-420F记录心电图的变化,以100 μmol/L利多卡因为阳性对照,采用全细胞膜片钳技术测定不同浓度(24.0、8.0、2.4、0.8 g/L)定悸复脉汤浓缩液对CHO细胞Nav1.5电流的影响,绘制量效曲线图并拟合IC50曲线。结果 模型组室性心律失常发生率为100.00%,滋阴潜阳养血安神组室性心律失常发生率为90.00%,对照组室性心律失常发生率为80.00%。滋阴潜阳养血安神组和对照组心律失常出现时间长于模型组,心律失常持续时间短于模型组,差异有统计学意义(P<0.05)。滋阴潜阳养血安神组和对照组心律失常出现时间、心律失常持续时间比较,差异无统计学意义(P>0.05)。采用贴壁培养方式,镜下可见CHO细胞成纤维细胞样,胞体较小,呈长梭形,为多突的纺锤形或星形的扁平细胞,细胞轮廓清楚,具有突起。100 μmol/L的利多卡因对CHO细胞Nav1.5电流的抑制率为(39.14±3.96)%。不同浓度(24.0、8.0、2.4、0.8 g/L)的定悸复脉汤浓缩药液对CHO细胞Nav1.5电流均有不同程度的抑制作用,抑制率分别为(48.79±0.65)%、(36.24±0.92)%、(26.95±1.80)%和(21.52±1.65)%,IC50=(24.0±2.4)g/L。结论 滋阴潜阳养血安神法可推迟室性心律失常出现时间,缩短室性心律失常持续时间,其作用可能与抑制Nav1.5电流有关。
Abstract:
Objective To investigate the effects of n Ziyin Qianyang Yangxue Anshen methodon electrocardiogram and Nav1.5 current in ventricular arrhythmia rats. Methods 30 healthy male SD rats were randomly divided into a model group, a control group and a Ziyin Qianyang Yangxue Anshen group, with 10 rats in each group. The model group was administered with saline, the control group was administered with metoprolol, the Ziyin Qianyang Yangxue Anshen group was administered with Dingji Fumai Decoction, and 2 weeks later, it was recorded by the BL-420F Electrocardiogram changes, 100 μmol/L lidocaine as a positive control, using whole-cell patch-clamp technology to determine different concentrations (24.0, 8.0, 2.4, 0.8 g/L) of Dingji Fumai Decoction for CHO cells Nav1.5 current effect, draw a dose-response curve and fit the IC50 curve.Results The rate of ventricular arrhythmias in the model group was 100.00%, the rate of ventricular arrhythmias in the Ziyin Qianyang Yangxue Anshen group was 90.00%, and the rate of ventricular arrhythmic rats in the control group It is 80.00%. The arrhythmia occurred longer in the Ziyin Qianyang Yangxue Anshen group and the control group than in the model group, and the arrhythmia duration was shorter than that in the model group,the difference was statistically significant (P<0.05). There was no significant difference in the occurrence time and duration of arrhythmia between the Ziyin Qianyang Yangxue Anshen group and the control group (P>0.05). Adherent culture was used to show that CHO cells were fibroblast-like, with small cell bodies and long spindle-shaped cells. They were spindle-shaped or star-shaped flat cells with multiple protrusions, with clear cell outlines and protrusions. The inhibition rate of 100 μmol / L lidocaine on Nav1.5 current in CHO cells was (39.14±3.96)%. Different concentrations (24.0, 8.0, 2.4, 0.8 g/L) of Dingji Fumai Decoction can inhibit Nav1.5 current in CHO cells to varying degrees. The inhibitory rates of Dingji Fumai Decoction concentrated liquid of different concentrations (24.0, 8.0, 2.4, 0.8 g/L) on Nav1.5 current in CHO cells were (48.79±0.65)%, (36.24± 0.92)%, (26.95±1.80)% and (21.52±1.65)%, IC50=(24.0 ± 2.4) g/L.Conclusion The method of Ziyin Qianyang Yangxue Anshencan delay the occurrence time of ventricular arrhythmia and shorten the duration of ventricular arrhythmia. Its effect may be related to the suppression of Nav1.5 current.
