摘要:
目的 探讨清窍胶囊对分泌性中耳炎大鼠Th17、Treg细胞及炎性因子的影响。方法 30只雌性SD大鼠,将其中24只构建分泌性中耳炎大鼠模型,造模成功后,24只大鼠随机分为模型组、强的松组(0.003 g/100 ml)、清窍胶囊低剂量治疗组(0.36 g/100 ml)、清窍胶囊高剂量治疗组(0.72 g/100 ml),每组6只,另6只设为对照组。治疗组分别按不同剂量灌胃给药,模型组与对照组灌服相同容积的蒸馏水,1次/d,连续14 d。ELISA检测血清IL-4、TGF-β、IL-6、IFN-γ、IL-10、IL-17水平,流式细胞技术检测脾脏和胸腺Th17、Treg细胞含量并比较。结果 与对照组比较,模型组大鼠TGF-β、IL-6、IFN-γ、IL-17、Th17、Treg细胞水平升高,IL-10、IL-4水平降低(P<0.05);与模型组比较,清窍胶囊高剂量治疗组TGF-β、IL-6、IFN-γ、IL-17、Th17、Treg细胞水平降低,IL-10、IL-4水平升高(P<0.05)。结论 清窍胶囊可有效调节Treg和Th17细胞介导的中耳炎的发生及发展,改善分泌性中耳炎。
Abstract:
Objective To investigate the effect of Qingqiao Capsule on Th17, Treg cells and inflammatory factors in rats with secretory otitis media. Methods 30 female SD rats, of which 24 were constructed as a rat model of secretory otitis media.After successful modeling, 24 rats were randomly divided into model group, prednisone group (0.003 g/100 ml), Qingqiao capsule low-dose treatment group (0.36 g/100 ml), and Qingqiao capsule high-dose treatment group (0.72 g/100 ml),There are 6 in each group, and the other 6 are set as control group.The treatment group was given different doses by intragastric administration. The model group and the control group were given the same volume of distilled water, once a day, for 14 consecutive days.ELISA was used to detect serum levels of IL-4, TGF-β, IL-6, IFN-γ, IL-10, and IL-17, and flow cytometry was used to detect and compare the contents of Th17 and Treg cells in the spleen and thymus.Results Compared with the control group, the levels of TGF-β, IL-6, IFN-γ, IL-17, Th17, and Treg cells in the model group increased, while the levels of IL-10 and IL-4 decreased (P<0.05);Compared with the model group, the levels of TGF-β, IL-6, IFN-γ, IL-17, Th17, and Treg cells in the Qingqiao capsule high-dose treatment group decreased, and the levels of IL-10 and IL-4 increased (P<0.05).Conclusion Qingqiao capsule can effectively regulate the occurrence and development of otitis media mediated by Treg and Th17 cells, and improve secretory otitis media.
