目的 运用孟德尔随机化(MR)的研究方法推断炎症性肠病(IBD)与牙周炎(PD)的因果关系。方法 收集欧洲牙周病学会(EFP)和国际炎症性肠病遗传学联盟(IBDGC)的大型全基因组关联研究(GWAS)数据,选择2个与牙周炎相关的单核苷酸多态性(SNPs)作为工具变量,分别是rs12641205和rs2668826。采用两样本MR方法分析牙周炎与IBD的因果关系,使用PLINK软件对PRISM和IBDGC数据进行质量控制和预处理,使MR-egger方法估计牙周炎对IBD的因果效应,使用Cochran’s Q检验和I2统计量来评估不同SNPs对因果效应的一致性。结果 牙周炎与IBD之间存在因果关系,牙周炎增加了IBD的发病风险(OR=1.18,95%CI:1.09~1.28,P<0.001)。不同SNPs对因果效应的估计结果基本一致(Q=0.41,P=0.52,I2=0)。结论 孟德尔随机化研究证实了牙周炎与炎症性肠病之间存在因果关联,为深入理解这两种疾病的发病机制和预防策略提供了新的视角。
Abstract:
Objective To investigate the causal relationship between inflammatory bowel disease (IBD) and periodontitis (PD) by using Mendelian randomization (MR). Methods The large-scale genome-wide association study (GWAS) data of the European Federation of Periodontology (EFP) and the International Inflammatory Bowel Disease Genetics Consortium (IBDGC) were collected. Two single nucleotide polymorphisms (SNPs) associated with periodontitis were selected as instrumental variables, namely rs12641205 and rs2668826. A two-sample MR method was used to analyze the causal relationship between periodontitis and IBD. PLINK software was used to perform quality control and preprocessing of PRISM and IBDGC data. The MR-egger method was used to estimate the causal effect of periodontitis on IBD. The Cochran’s Q test and I2 statistics were used to evaluate the consistency of different SNPs on the causal effect. Results There was a causal relationship between periodontitis and IBD, and periodontitis increased the risk of IBD (OR=1.18, 95%CI: 1.09-1.28, P<0.001). The estimation results of causal effect of different SNPs were basically consistent (Q=0.41, P=0.52, I2=0). Conclusions The Mendelian randomization study confirmed the causal association between periodontitis and inflammatory bowel disease, providing a new perspective for understanding the pathogenesis and prevention strategies of these two diseases.
