目的 观察在脑缺血/再灌注损伤大鼠脑组织中不同时间点HO-1/HO-1mRNA的表达趋势,为脑缺血/再灌注损伤的机制研究与防治提供了一定的理论依据。方法 成年雄性SD大鼠25只随机分为五组:假手术组,脑缺血2 h再灌注3 h组,6 h组,12 h组和24 h组,每组5只。各组大鼠分别于再灌注后相应时间点取缺血侧大脑皮质进行RT-qPCR检测,对HO-1基因mRNA的表达进行定量分析;采用Western blotting方法检测各时间点HO-1蛋白表达。结果 假手术组HO-1基因mRNA表达水平较低,与之相比脑缺血/再灌注3 h时升高(P<0.05),再灌注6 h达到高峰(P<0.05),与6 h相比再灌注12 h时降低(P<0.05),与再灌注24 h接近但仍高于再灌注3 h时水平(P<0.05);HO-1蛋白在缺血再灌注各个时间点表达规律与HO-1基因mRNA表达水平一致。结论 脑缺血/再灌注损伤后大鼠缺血脑组织中HO-1/HO-1mRNA表达水平于损伤早期(6 h)迅速升高,达高峰后又逐渐下降并保持在一定水平,是脑缺血/再灌注损伤早期重要机制之一。
Abstract:
Objective To observe the expression of HO-1/HO-1 mRNA at different time points in rats with cerebral ischemia/reperfusion injury,and provide a theoretical basis for the study and prevention of cerebral ischemia/reperfusion injury.Methods 25 adult male Sprague-Dawley rats were randomly divided into five groups:sham operation group,cerebral ischemia 2 h reperfusion 3 h group,6 h group,12 h group and 24 h group,5 rats in each group.Rats in each group were examined by RT-qPCR at the corresponding time points after reperfusion.The expression of HO-1 gene mRNA was quantitatively analyzed.Western blotting was used to detect HO-1 protein expression at each time point.Results The expression level of HO-1 gene mRNA was lower in sham operation group.In contrast,cerebral ischemia/reperfusion increased at 3 h(P<0.05),reached the peak at 6 h(P<0.05),decreased at 12 h after reperfusion compared with 6 h(P<0.05),and was close to 24 h after reperfusion,but still higher than that at 3 h after reperfusion(P<0.05).The expression of HO-1 protein was consistent with the expression of HO-1 gene mRNA at different time points after ischemia reperfusion.Conclusion The expression of HO-1/HO-1mRNA in the ischemic brain tissue of rats after cerebral ischemia/reperfusion injury increased rapidly at the early stage of injury(6 h),then decreased gradually and maintained at a certain level after reaching the peak,which is one of the important mechanisms in the early stage of cerebral ischemia /reperfusion injury.
