摘要:
目的 分析甲状旁腺激素应用于大鼠脊髓损伤后继发骨质疏松症的治疗效果。方法 购买20只SD大鼠,雌雄各半,建立脊髓损伤(SCI)动物模式,检测全部大鼠骨密度(BMD),选取12只BMD偏低的大鼠并随机分为对照组与观察组,对照组大鼠不予骨质疏松治疗,观察组大鼠给予甲状旁腺激素(PTH)注射;观察两组在应用PTH后1周、3 周、6 周、12周四个监测时间点时的股骨、胫骨骨密度(BMD),骨钙素(OCN),碱性磷酸酶(ALP)的各项指标的检测结果并进行对比分析。结果 建模术后6 d时两组大鼠BMD均显著低于本组术前(P<0.01),但两组间无统计学差异(P>0.05);能够满足本次研究要求。观察组胫骨、股骨BMD多时间点监测结果显著高于对照组,具有统计学意义(P<0.01);自第6~12周时BMD出现显著上升趋势。观察组OCN多时间点监测结果显著高于对照组(P<0.01);OCN表达在第1~6周时呈上升趋势,于第6 周达峰值后开始回落,第6 ~12周时可见明显下降。观察组ALP多时间点监测结果显著高于对照组(P<0.01);ALP表达水平自第1周起呈现出上升趋势,第6 ~12周上升最为显著。结论 甲状旁腺激素对于脊髓损伤后继发的骨质疏松的大鼠具有明确的治疗效果,据此可以认为脊髓损伤早期应用甲状旁腺激素能够在一定程度上预防继发骨质疏松的发生。
Abstract:
Abstract:Objective To analyze the therapeutic effect of parathyroid hormone(PTH)on osteoporosis leaded by spinal cord injury in rats.Methods 20 SD rats,half of which are males and the other half females,were purchased.An animal model of spinal cord injury(SCI)was established and bone mineral density(BMD)was measured in all rats.12 rats with low BMD were randomly divided into two groups:the control group and the observation group.Rats in the control group were not treated with osteoporosis,and the rats in the observation group were injected with parathyroid hormone(PTH). The bone mineral density(BMD)and osteocalcin(OCN)of the femur and tibia were observed in 1 week,3 weeks,6 weeks and 12 weeks after PTH.The results of alkaline phosphatase(ALP)were compared and analyzed.Results 6 d after modeling,BMD of both two groups were significantly lower than that in the group before operation,but there was no statistical difference between the two groups,P>0.05,able to meet the requirements of this study. The BMD in the tibia and femur in the observation group were significantly higher than those in the control group at multiple time points,P<0.01 having statistical significance.The BMD obviously increased in the first 6 to 12 weeks.The observation group OCN monitoring results was significantly higher than the control group at multiple time points P<0.01;the expression of OCN increased in the first 1 to 6 weeks,and decreased significantly from the 6th to 12th week after reaching a peak at the 6 w.The observation group ALP was significantly higher than the control group P<0.01;The expression of ALP showed an increasing trend since the first week.At week 6 and 12, the most significant increase was observed.Conclusion Parathyroid hormone has a definite therapeutic effect on osteoporosis rats after spinal cord injury.It is concluded that parathyroid hormone can prevent secondary osteoporosis in the early stage of spinal cord injury.
