摘要:
目的 研究黄芪对细胞毒素相关蛋白A(cagA)诱导大鼠的系膜细胞外基质分泌的影响。方法 将大鼠系膜细胞进行体外培养,并随机分为四组,分别为空白对照组、IL-1β阳性对照组(10 ng/ml IL-1β)和CagA实验组(4 μg/ml CagA)及CagA+黄芪干预组(1 mg/ml的黄芪注射液+4 μg/ml CagA),各组予以对应处理后刺激培养72 h,检测细胞培养上清液Ⅰ型胶原(CollagenⅠ)、Ⅲ型胶原(CollagenⅢ型)蛋白水平及其mRNA表达。结果 IL-1β阳性对照组、CagA实验组CollagenⅠ、CollagenⅢ蛋白浓度及mRNA表达水平高于空白对照组,CagA+黄芪干预组CollagenⅠ、CollagenⅢ蛋白浓度及mRNA表达水平低于CagA实验组,差异有统计学意义(P<0.05)。结论 黄芪或通过减少Ⅰ、Ⅲ型胶原蛋白的分泌,抑制CagA诱导的大鼠系膜细胞外基质的异常分泌,可为IgA肾病的治疗提供思路。
Abstract:
Objective To investigate the effects of Astragalus Membranaceus on CagA-induced extracellular matrix secretion of rat mesangial cells.Methods Rat mesangial cells were cultured in vitro and randomly divided into four groups: blank control group, IL-1β positive control group (10 ng/ml IL-1β), CagA experimental group (4 μg/ml CagA) and CagA+Astragalus Membranaceus intervention group (1 mg/ml Astragalus Membranaceus injection+4 μg/ml CagA). Each group was stimulated for 72 h after corresponding treatment. The protein level and mRNA expression of type Ⅰ collagen (CollagenⅠ) and type Ⅲ collagen (CollagenⅢ) in cell culture supernatant were detected.Results The protein concentration and mRNA expression of CollagenⅠ and CollagenⅢin IL-1β positive control group and CagA experimental group were higher than those in blank control group, and the protein concentration and mRNA expression of CollagenⅠ and CollagenⅢ in CagA+Astragalus Membranaceus intervention group were lower than those in CagA experimental group, the difference was statistically significant (P<0.05).Conclusion Astragalus Membranaceus can inhibit CagA-induced abnormal secretion of extracellular matrix in rat mesangial cells by reducing the secretion of type Ⅰ and type Ⅲ collagen, which can provide ideas for the treatment of IgA nephropathy.
