摘要:
目的 探讨硫化氢(H2S)对胃癌多药耐药性(MDR)的影响。方法 采用逐步递增顺铂浓度法建立胃癌多药耐药细胞株MGC803/MDR,显微镜观察细胞形态,CCK8法检测细胞对顺铂、阿霉素、5-氟尿嘧啶的IC50值,亚甲基蓝分光光度计法检测H2S含量,Western blot检测胱硫醚-β-合成酶(CBS)表达。结果 MGC803/MDR细胞在显微镜下排列松散,胞体较小,细胞形态呈梭形或不规则形。MGC803/MDR细胞对顺铂、阿霉素、5-氟尿嘧啶的IC50值显著高于MGC803细胞(P<0.01);MGC803/MDR细胞H2S、CBS表达显著高于MGC803细胞(P<0.01);采用氨基氧乙酸(AOAA)作用24 h后,MGC803/MDR细胞H2S生成显著减少(P<0.01),对顺铂、阿霉素、5-氟尿嘧啶的IC50值显著降低(P<0.01)。采用NaSH作用24 h后,MGC803细胞对顺铂、阿霉素、5-氟尿嘧啶的IC50值显著升高(P<0.01)。结论 气体信号分子H2S参与了胃癌多药耐药性的形成。
Abstract:
Objective To investigate the effect of hydrogen sulfide (H2S) on multidrug resistance of gastric cancer.Methods Multidrug resistant gastric cancer cell line MGC803/MDR were established by stepwise dose escalation with cisplatin. Cell morphology was observed under an optical microscope. Cell sensitivity to cisplatin, adriamycin and 5-fluorouracil was analyzed by computing IC50 value which were determined by CCK8 assay. The methylene blue assay was used to detecte H2S. The protein level of cystathionine β synthase (CBS) was detected by Western blot analysis.Results MGC803/MDR cells were loosely arranged with small cell bodies and spindle or irregular cell morphology under microscope observation. The IC50 values of cisplatin, adriamycin and 5-fluorouracil in MGC803/MDR cells were significantly higher than in MGC803 cells (P<0.01). The level of H2S and the protein level of CBS in MGC803/MDR cells were significantly higher than in MGC803 cells (P<0.01). After 24 h treatment with aminoxyacetic acid (AOAA), the production of H2S in MGC803/MDR cells was significantly reduced, and the IC50 values of cisplatin, adriamycin and 5-fluorouracil in MGC803/MDR cells were significantly decreased (P<0.01). After 24 h treatment with NaSH, the IC50 values of cisplatin, adriamycin and 5-fluorouracil in MGC803 cells were significantly increased (P<0.01).Conclusion H2S mediates multidrug resistance of gastric cancer.
