摘要:
目的 通过构建“药物-成分-靶点-疾病”的相互作用网络,探究脊蛇祛湿胶囊中的君药金毛狗脊治疗类风湿性关节炎(RA)的机制。方法 通过CNKI和Pubchem数据库查找金毛狗脊的活性成分并用Swiss Target Prediction数据库筛选得到的靶点,通过GeneCards和omim数据库整合筛选得到的靶点,将其取交集得到共同靶点导入Cytoscape行进可视化分析,并通过String和Cytoscape进行蛋白质-蛋白质相互作用网络分析,运用Metescape数据库将得到的结果进行GO和KEGG富集分析。结果 通过Swiss Target Prediction得到金毛狗脊167个药物靶点,GeneCards和omim数据库整合筛选RA 3060个靶点,取其交集共91个共同基因,通过String蛋白互作网络筛选得到20个关键基因;GO分析显示金毛狗脊涉及炎症反应等生物过程;KEGG分析显示金毛狗脊参与调节EGFR酪氨酸激酶抑制剂耐药性通路、HIF-1信号通路、PI3K-AKT信号通路、雌激素信号通路等。结论 金毛狗脊的药效成分与类风湿性关节炎有相同的作用靶点,与其相关的靶点包括ALB、MMP9、EGFR等,金毛狗脊可以通过调节多种通路抑制类风湿性关节炎的进程。
Abstract:
Objective To explore the mechanism of Cibotium barometz of Jishe Qushi capsule in the treatment of rheumatoid arthritis(RA) by constructing the interaction network of “drug-component-target-disease”.Methods Through the CNKI and Pubchem database to find the active ingredients of Cibotium barometz, and using the Swiss Target Prediction database to screen the targets, through the GeneCards and omim database integration to screen the targets, the common targets obtained by the intersection were imported into Cytoscape for visual analysis, and the protein-protein interaction network analysis was carried out by String and Cytoscape. The results were analyzed by GO and KEGG enrichment analysis using Metescape database.Results A total of 167 drug targets were obtained by Swiss Target Prediction, and 3060 RA targets were screened by GeneCards and omim database integration. A total of 91 common genes were intersected, and 20 key genes were screened by String protein interaction network. GO analysis showed that the Cibotium barometz involved biological processes such as inflammatory response. KEGG analysis showed that Cibotium barometz was involved in the regulation of EGFR tyrosine kinase inhibitor resistance pathway, HIF-1 signaling pathway, PI3K-AKT signaling pathway, estrogen signaling pathway and so on.Conclusion The active ingredients of Cibotium barometz have the same target as rheumatoid arthritis, the related targets include ALB, MMP9, EGFR, etc. Cibotium barometz can inhibit the process of rheumatoid arthritis by regulating multiple pathways.
