摘要:
目的 基于网络药理学探析芙蓉尿感清治疗尿路感染的潜在靶点、信号通路及作用机制。方法 通过TCMSP数据库及文献检索获取芙蓉尿感清的药物活性成分及作用靶点,并借助Uniprot数据库校正基因名称;通过Genecards、DisGeNET数据库收集治疗尿路感染的相关靶点,利用Venny2.1获取药物-疾病交集靶点;利用STRING数据库构建交集靶点的蛋白质-蛋白质相互作用(PPI)网络,运用软件Cytoscape对核心靶点进行拓扑分析;采用DAVID数据库对核心靶点进行GO及KEGG富集分析;运用Cytoscape3.9.1软件构建“药物-活性成分-靶点-疾病-通路”网络图。结果 筛选得到芙蓉尿感清活性成分共145个,主要为槲皮素、木犀草素、黄芩素等黄酮类化合物,靶点基因281个;收集疾病靶点10 308个,交集靶点262个;PPI蛋白网络及拓扑分析发现AKT1、TP53、TNF等关键靶点;GO富集分析显示,生物学过程主要涉及基因表达、细胞增殖及凋亡过程、炎症反应等;细胞组分涉及细胞外间隙、质膜、核质等;分子功能涉及ATP结合、蛋白激酶结合、DNA结合等;KEGG富集分析获得147条通路,主要涉及PI3K/Akt、Mark信号通路。结论 芙蓉尿感清治疗尿路感染具有多成分、多途径、多靶点的特点,其作用机制可能是通过介导AKT1、TP53、TNF、IL6等关键靶点的表达,调控PI3K/Akt、Mark信号通路,发挥抗炎、抑制菌群活性、调节机体免疫等作用,从而消除尿路炎症和改善临床症状。
Abstract:
Objective To explore the potential targets, signaling pathways and mechanism of FurongNiaoganqing in the treatment of urinary tract infection based on network pharmacology.Methods The active components and targets of Furong Niaoganqing were obtained by TCMSP database and literature retrieval, and the gene name was corrected by Uniprot database. Genecards and DisGeNET databases were used to collect relevant targets for the treatment of urinary tract infection, and Venny2.1 was used to obtain drug-disease intersection targets. The protein-protein interaction (PPI) network of intersection targets was constructed by STRING database, and the topological analysis of core targets was carried out by Cytoscape software. GO and KEGG enrichment analysis of core targets was performed using DAVID database. Cytoscape3.9.1 software was used to construct a “drug-active ingredient-target-disease-pathway” network diagram.Results A total of 145 active components of Furong Niaoganqing were screened, mainly flavonoids such as quercetin, luteolin and baicalein, and 281 target genes were identified. A total of 10 308 disease targets and 262 intersection targets were collected. PPI protein network and topology analysis found key targets such as AKT1, TP53 and TNF; GO enrichment analysis showed that biological processes mainly involved gene expression, cell proliferation and apoptosis, inflammatory response, etc;cell components involve extracellular space, plasma membrane, nucleoplasm, etc.; the molecular functions involved ATP binding, protein kinase binding, DNA binding, etc. KEGG enrichment analysis obtained 147 pathways, mainly involving PI3K/Akt and Mark signaling pathways.Conclusion Furong Niaoganqing has the characteristics of multi-component, multi-pathway and multi-target in the treatment of urinary tract infection. Its mechanism may be to regulate PI3K/Akt and Mark signaling pathways by mediating the expression of key targets such as AKT1, TP53, TNF and IL6, so as to exert anti-inflammatory, inhibit bacterial activity and regulate body immunity, thereby eliminating urinary tract inflammation and improving clinical symptoms.
