摘要:
目的 确定高迁移率族蛋白B1与胃癌组织血管生成之间的联系,筛选胃癌诊断和治疗的靶点。方法 收集2008年6月~2016年6月怀化市第一人民医院病理科正常胃、胃癌旁、胃癌癌组织蜡块共105例,采用常规HE染色(苏木素-伊红)、免疫组织化学染色(链霉菌抗生物素蛋白-过氧化物酶连结法-SP法),检测正常胃组织、胃癌旁组织及胃癌组织中HMGB1的表达情况、检测人造血祖细胞抗原(CD34)的表达情况来反映正常胃组织、胃癌旁组织及胃癌组织中的微血管密度(MVD),统计学分析HMGB1的表达与胃癌组织MVD之间存在的相关性。结果 正常胃组织HMGB1的阳性表达率低,胃癌癌旁组织HMGB1的阳性表达率增高、胃癌组织中HMGB1的阳性表达率最高,在三种组织中HMGB1的表达存在差异(P<0.05);HMGB1的表达水平与MVD表现出正相关性(r=0.205,P<0.05)。结论 HMGB1在胃组织、胃癌旁组织、胃癌组织中的表达呈阶梯型增高,胃癌组织中HMGB1的表达水平与组织MVD成正相关性。
Abstract:
Abstract:Objective To determine the relationship between high mobility group protein B1 and angiogenesis in gastric carcinoma, and to screen the target of diagnosis and treatment of gastric carcinoma. Methods A total of 105 cases of normal stomach, gastric carcinoma and gastric carcinoma tissue were collected from June 2008 to June 2016 in the Department of Pathology, First People's Hospital of Huaihua City, using conventional HE staining (hematoxylin-eosin) and immunohistochemical staining. Streptomyces avidin-peroxidase-linked-SP method, detecting the expression of HMGB1 in normal gastric tissue, para-cancerous tissues and gastric carcinoma tissues, and detecting the expression of human hematopoietic progenitor antigen (CD34) to reflect normal Microvessel density (MVD) in gastric tissue, paracancerous tissues and gastric carcinoma tissues, and statistical analysis of the correlation between HMGB1 expression and MVD in gastric carcinoma tissues.Results The positive expression rate of HMGB1 in normal gastric tissue was low, the positive expression rate of HMGB1 in gastric carcinoma tissues was increased, and the positive expression rate of HMGB1 was the highest in gastric carcinoma tissues. The expression of HMGB1 was different in three tissues (P<0.05). The expression level of HMGB1 was positively correlated with MVD (r=0.205,P<0.05).Conclusion The expression of HMGB1 in gastric tissue, paracancerous tissues and gastric carcinoma tissues is increased stepwise. The expression level of HMGB1 in gastric carcinoma tissues is positively correlated with tissue MVD.
