摘要:
目的 研究丹酚酸B(Sal B)对D-半乳糖联合氯化铝诱导的阿尔茨海默病(AD)小鼠认知功能保护作用和机制。方法 将6月龄SPF级小鼠随机分为对照组、AD模型组、Sal B低剂量组(30 mg/kg)、Sal B高剂量组(60 mg/kg),每组10只,连续灌胃给药8周。采用跳台实验观察各组小鼠认知和记忆能力;比色法检测AD小鼠血清及海马的氧化指标超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)以及丙二醛(MDA);Western Blot 法检测半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)、B淋巴细胞瘤-2 基因(Bcl-2)、Bcl-2相关X蛋白(Bax)表达并比较。结果 Sal B各组较AD模型组认知功能及记忆水平改善,SOD和GSH-PX 活力升高、MDA 水平降低,Caspase-3、Bax、Bax/Bcl-2表达减少,Bcl-2表达增加。结论 Sal B对AD小鼠认知功能具有一定的保护作用,其可能的机制是改善氧化应激损伤,减少神经元凋亡。
Abstract:
Objective To study the protective effect and mechanism of salvianolic acid B (Sal B) on the cognitive function of Alzheimer’s disease (AD) mice induced by D-galactose combined with aluminum chloride.Methods 6-month-old SPF mice were randomly divided into control group, AD model group, Sal B low-dose group (30 mg/kg), and Sal B high-dose group (60 mg/kg), each group had 10 mice,they were administered intragastrically for 8 weeks.Using platform jumping experiment to observe the cognitive and memory abilities of mice in each group;Colorimetric detection of AD mouse serum and hippocampus oxidation indicators superoxide dismutase (SOD), glutathione peroxidase (GSH-PX) and malondialdehyde (MDA);Western Blot was used to detect the expression of Caspase-3 (Caspase-3), B lymphocyte tumor-2 (Bcl-2), and Bcl-2 related X protein (Bax) and compare them.Results Compared with the AD model group, the Sal B group improved cognitive function and memory level, SOD and GSH-PX activity increased, and MDA level decreased.The expression of Caspase-3, Bax, Bax/Bcl-2 decreased, and the expression of Bcl-2 increased.Conclusion SalB has a certain protective effect on the cognitive function of AD mice, and its possible mechanism is to improve oxidative stress damage and reduce neuronal apoptosis.
