摘要:
目的 探讨KIRREL表达在胃癌中的预后作用及与肿瘤免疫细胞浸润的相关性。方法 通过TIMER和UALCAN数据库分析KIRREL在泛癌和胃癌中的表达以及与临床特征的关系。Sangerbox和Kaplan-Meier数据库分析KIRREL表达对胃癌患者预后的影响。TIMER数据库分析KIRREL表达与免疫细胞浸润以及浸润等级与患者预后的关系。Sangerbox数据库分析KIRREL基因与免疫检查点基因以及ESTIMATE分数的关系。结果 KIRREL在胆管癌、食管癌、头颈鳞状细胞癌、胃癌中的表达高于相对应的癌旁组织(P<0.05),且在膀胱尿路上皮癌、乳腺浸润癌、肾嫌色细胞癌、肾乳头状细胞癌、肺腺癌、肺鳞癌、前列腺癌、甲状腺癌、子宫内膜癌中表达下调(P<0.05)。通过UALCAN数据库分析显示,KIRREL在胃癌组织中表达具有显著差异(P=1.18E-06)。UALCAN数据库分析显示,KIRREL表达与肿瘤分期、分级、淋巴结转移分期有关。GEPIA数据库分析显示,高表达的KIRREL对胃癌患者OS有显著影响(HR=1.5,P=0.02)。Kaplan-Meier数据库分析显示,更高表达的KIRREL预示着胃癌患者具有较差的OS(HR=1.5,P=2.7E-06)。TIMER数据库分析显示,KIRREL表达与CD8+T细胞、CD4+T细胞、巨噬细胞、中性粒细胞、树突状细胞的浸润等级呈正相关(P<0.05),与肿瘤纯度、B细胞无相关性(P>0.05)。KIRREL的表达与基质分数、免疫分数和评估分数均呈正相关。结论 KIRREL在胃癌的发生、发展及免疫细胞浸润中发挥了关键作用,可以作为评估患者临床预后的生物学标记物和指导免疫治疗的潜在靶点。
Abstract:
Objective To investigate the prognostic role of KIRREL expression in gastric cancer and its correlation with tumor immune cell infiltration.Methods The expression of KIRREL in pan-cancer and gastric cancer and its relationship with clinical features were analyzed by TIMER and UALCAN databases. Sangerbox and Kaplan-Meier databases were used to analyze the effect of KIRREL expression on the prognosis of patients with gastric cancer. TIMER database was used to analyze the relationship between KIRREL expression and immune cell infiltration, as well as the relationship between infiltration grade and prognosis of patients. The relationship between KIRREL gene and immune checkpoint gene and ESTIMATE score was analyzed by Sangerbox database.Results The expression of KIRREL in cholangiocarcinoma, esophageal cancer, head and neck squamous cell carcinoma and gastric cancer was higher than that in corresponding adjacent tissues (P<0.05), and the expression of KIRREL was down-regulated in bladder urothelial carcinoma, breast invasive carcinoma, chromophobe cell carcinoma, papillary renal cell carcinoma, lung adenocarcinoma, lung squamous cell carcinoma, prostate cancer, thyroid cancer and endometrial carcinoma (P<0.05). UALCAN database analysis showed that KIRREL expression was significantly different in gastric cancer tissues (P=1.18E-06). UALCAN database analysis showed that KIRREL expression was related to tumor stage, grade and lymph node metastasis stage. GEPIA database analysis showed that high expression of KIRREL had a significant effect on OS of gastric cancer patients (HR=1.5, P=0.02). Kaplan-Meier database analysis showed that higher expression of KIRREL indicated poor OS in gastric cancer patients (HR=1.5, P=2.7E-06). TIMER database analysis showed that KIRREL expression was positively correlated with the infiltration grade of CD8+ T cells, CD4+ T cells, macrophages, neutrophils and dendritic cells (P<0.05), but not with tumor purity and B cells (P>0.05). The expression of KIRREL was positively correlated with matrix fraction, immune fraction and evaluation score.Conclusion KIRREL plays a key role in the occurrence, development and immune infiltration of gastric cancer, and can be used as a biological marker to evaluate the clinical prognosis of patients and a potential target to guide immunotherapy.
