摘要:
目的 基于网络药理学探讨桃红大将逐瘀汤加减治疗便秘的作用机制。方法 采用TCMSP数据库检索和筛选桃红大将逐瘀汤加减的中药活性化合物及其靶点;运用GeneGards和OMIM数据库筛选出与便秘相关的基因,将其与药物靶点交集获得靶基因并构建活性化合物-靶基因网络。利用String数据库构建靶基因的PPI网络,并筛选出关键基因;再对靶基因进行GO和KEGG富集分析。结果 桃红大将逐瘀汤加减有82个活性化合物作用于227个基因,与疾病相关的基因有4663个,通过交集得到177靶基因;其中ALB、AKT1、IL6、JUN、CASP3、VEGFA、IL1B、EGFR、PTGS2、MYC等10个基因是关键基因,主要富集在169个GO条目以及IL-17信号通路、TNF信号通路等158个KEGG通路中。结论 桃红大将逐瘀汤加减主要通过ALB、AKT1、IL6等基因及IL-17信号通路和TNF信号通路调节肠道蠕动功能,达到治疗便秘目的。
Abstract:
Objective To investigation the mechanism of modified Taohong Dajiang Zhuyu decoction in the treatment of constipation based on network pharmacology.Methods TCMSP database was used to search and screen the active compound and target of modified Taohong Dajiang Zhuyu decoction. Genes related to constipation were screened by GeneGards and OMIM database, and target genes were obtained by intersection with drug targets, and active compound target gene network was constructed. PPI network of target genes was constructed using String database, and key genes were selected out. GO and KEGG enrichment analysis was done for target genes.Results There were 82 active compounds acting on 227 genes, 4663 genes related to disease, 177 target genes were delivered by intersection. Ten genes including ALB, AKT1, IL6, JUN, CASP3, VEGFA, IL1B, EGFR, PTGS2 and MYC were key genes, which were mainly enriched in 169 GO items and 158 KEGG pathways including IL-17 signaling pathway and TNF signaling pathway.Conclusion Modified Taohong Dajiang Zhuyu decoction mainly regulates intestinal peristalsis through ALB, AKT1, IL6 and OTHER genes as well as IL-17 signaling pathway and TNF signaling pathway, thus achieving the objective of treating constipation.
