摘要:
目的 通过观察Claudin-14在两种实验性大鼠肾结石模型中表达的差异,探究纳米细菌在结石形成过程中的作用。方法 选取90只Wistar雄性大鼠,随机分为纳米细菌组(NB组,大鼠尾静脉注射NB+生理盐水灌胃)、乙二醇组(EG组,大鼠1%乙二醇饮水和2%氯化铵AC 2 ml灌胃2周)、空白对照组(NC组,大鼠尾静脉注射生理盐水+生理盐水灌胃),人工采集大鼠血、尿进行钙浓度检测,取大鼠肾脏组织行结石成分分析及免疫组化检测Claudin-14改变。结果 NB组、EG组含草酸钙结石分别占总结石数的61.54%、78.95%;NB组与EG组1~10周内血钙、尿钙含量分别比较,差异无统计学意义(P>0.05);EG组第3周后Claudin-14蛋白的表达显著增强,NB组第4周后Claudin-14蛋白的表达开始增强,EG组Claudin-14蛋白的表达显著强于NB组,但NC组Claudin-14蛋白的表达未见明显改变。结论 纳米细菌是形成结石的原因之一,草酸钙结晶的形成不仅有纳米细菌的参与,同时Claudin-14所参与的细胞通路也起到了相应的作用,导致尿钙升高,最终形成结石。
Abstract:
Objective To investigate the role of nanobacteria in the formation of renal calculi by observing the difference of Claudin-14 expression in two experimental rat renal calculi models.Methods A total of 90 male Wistar rats were randomly divided into nanobacteria group (NB group, rats were intragastrically administrated with NB+normal saline via tail vein), ethylene glycol group (EG group, rats were intragastrically administrated with 1% ethylene glycol drinking water and 2% ammonium chloride AC 2 ml for 2 weeks), and blank control group (NC group, rats were intragastrically administrated with normal saline via tail vein+normal saline). The blood and urine of rats were collected artificially for calcium concentration detection. The kidney tissues of rats were taken for stone composition analysis and immunohistochemical detection of Claudin-14 changes.Results Calcium oxalate stones in NB group and EG group accounted for 61.54% and 78.95% of the total stones, respectively. There was no significant difference in blood calcium and urine calcium between NB group and EG group within 1-10 weeks (P>0.05). The expression of Claudin-14 protein in EG group was significantly increased after 3 weeks, and the expression of Claudin-14 protein in NB group began to increase after 4 weeks; the expression of Claudin-14 protein in EG group was significantly stronger than that in NB group, but there was no significant change in NC group.Conclusion Nanobacteria are one of the reasons for the formation of stones. The formation of calcium oxalate crystals is not only involved in nanobacteria, but also in the cell pathway involved in Claudin-14, leading to the increase of urinary calcium and the formation of stones.
