摘要:
目的 探讨骨关节炎(OA)与2型糖尿病(T2DM)风险的因果关联。方法 利用来自不同研究机构(如arcOGEN、UKBiobank)的全基因组关联研究(GWAS)汇总数据,以OA作为暴露因素,T2DM作为结局变量。采用多重孟德尔随机化(MR)分析方法,包括反方差加权法(IVW)、加权中位数法(WM)、MR-Egger法、多效性检验和异质性检验等,评估OA与T2DM风险之间的关联性。结果 IVW法结果显示,OA和T2DM的高风险相关(OR=1.002,95%CI:1.000~1.003,P=0.012);MR-Egger、WM的OR值与IVW的OR值的方向一致;MR-Egger回归显示,与OA相关的SNPs均不具有水平多向性(P>0.05)。Cochrane’s Q检验显示在IVs中不存在异质性(P>0.05);敏感性分析显示因果效应稳定。结论 MR分析支持OA与T2DM风险增加有关。
Abstract:
Objective To explore the causal association between osteoarthritis(OA) and the risk of type 2 diabetes mellitus(T2DM). Methods Genome-wide association study (GWAS) summary data from different research institutions (such as arcOGEN, UKBiobank) were used. OA was used as an exposure factor and T2DM was used as an outcome variable. Multiple Mendelian randomization (MR) analysis methods, including inverse variance weighting (IVW), weighted median (WM), MR-Egger method, pleiotropic test and heterogeneity test, were used to evaluate the association between OA and T2DM risk. Results The results of IVW showed that OA was associated with high risk of T2DM (OR=1.002, 95%CI: 1.000-1.003, P=0.012). The OR value of MR-Egger and WM was consistent with that of IVW. MR-Egger regression showed that the SNPs associated with OA did not have horizontal polytropy (P>0.05). Cochrane’s Q test showed no heterogeneity in IVs (P>0.05). Sensitivity analysis showed that the causal effect was stable. Conclusion MR analysis support that OA is associated with an increased risk of T2DM.
