摘要:
目的 研究米诺环素对脑出血大鼠VEGF、GLUT-1表达的影响,探讨其的脑保护作用机制。方法 选用雄性Wistar大鼠60只,随机分为假手术组、模型组及米诺环素组,每组20只。采用Ⅶ型胶原酶法制备Wistar大鼠右侧纹状体出血模型。模型制备成功后米诺环素组给予腹腔注射米诺环素,剂量为22.5 mg/kg,2次/d,假手术组和模型组给予腹腔注射等量的生理盐水。观察脑缺血后改良神经功能缺损评分(mNSS)、脑组织含水量,Western blots及免疫组化法分别检测VEGF及GLUT-1的表达。结果 与模型组相比,米诺环素组大鼠脑组织的mNSS评分、脑组织含水量、VEGF及GLUT-1的表达量降低,差异均有统计学意义(P<0.05)。结论 米诺环素可显著减轻脑出血后大鼠的神经功能损害及组织学损伤,并提高大鼠脑组织VEGF及GLUT-1的表达水平,对脑出血具有一定的神经保护作用,其机制可能与增强VEGF及GLUT-1的表达有关。
Abstract:
Objective To investigate the effects of minocycline (MC) on the expression of VEGF and GLUT-1 in rats with intracerebral hemorrhage, and to explore the mechanism of brain protection of minocycline. Methods 60 male Wistar rats were randomly divided into sham operation group, model group and minocycline group, with 20 rats in each group. The right striatum hemorrhage model of Wistar rats was prepared by type VII collagenase method. After successful model preparation, the minocycline group was given intraperitoneal injection of minocycline at a dose of 22.5 mg/kg twice daily. The sham operation group and the model group were given an equal amount of normal saline. The cerebral ischemic modified neurological deficit score (mNSS), brain tissue water content, Western blots and immunohistochemistry were used to detect the expression of VEGF and GLUT-1.Results Compared with the model group, the mNSS score, brain water content, VEGF and GLUT-1 expression in the brain tissue of the minocycline group were significantly lower (P<0.05).Conclusion Minocycline can significantly reduce the neurological damage and histological damage in rats after intracerebral hemorrhage, and increase the expression of VEGF and GLUT-1 in rat brain tissue. It has a certain neuroprotective effect on intracerebral hemorrhage, and its mechanism may be associated with enhanced expression of VEGF and GLUT-1.
